چکیده
مقدمه: دگرگونی و تمایز سلولهای B فعال شده به پلاسموسیتها و همچنین، سلولهای خاطرهدار وابسته به پیامهای حاصل از گیرندهی سلول B میباشد. پیامهای ناشی از گیرندهی آنتیژن و گیرندههای سیتوکاینی سطح سلولهای B، سبب القای بروز عوامل رونوشتبرداری خاصی میشوند که در نهایت این عوامل، تعیین کنندهی سرنوشت سلول B میباشند.روشها: جداسازی سلولهای تک هستهای خون محیطی (Peripheral blood mononuclear cells یا PBMCs) با استفاده از گرادیان شیب غلظت و با استفاده از فایکول انجام گرفت و سپس، جداسازی سلولهای B خالص با روش Magnetic-activated cell sorting (MACS) انجام شد. در مرحلهی بعد، برای تحریک و تمایز سلولهای B به پلاسمابلاستها، این سلولها در محیط Roswell Park Memorial Institute1640 (RPMI1640) در حضور Purified anti- human CD40 antibody و Anti-IgM f(ab)`2 یا و Purified anti-human CD40 Antibody و لیپوپلیساکارید (Lipopolysaccharides یا LPS) کشت داده و سپس، پلاسمابلاستها با استفاده از سه نشانگر CD38+، CD27+ و IgM- به روش فلوسایتومتری ارزیابی شد.یافتهها: در محیط In vitro، با تحریک دایمی لنفوسیتهای B از طریق Cross-link کردن گیرندهی آنها (B cell receptor یا BCR) و تحریک با Anti-human CD40 antibody و Anti-IgM f(ab)`2 و یا Anti-CD40 و LPS اغلب سلولهای B زنده مانده بودند و حتی تمایز آنها به ردهی دیگری از سلولهای B (پلاسمابلاستهای CD38+، CD27+ و IgM-) مشاهده شد. از نظر آماری، تفاوت معنیداری در بیان نشانگرهای پلاسمابلاستی در سطح سلولها در هر دو حالت تحریکی مشاهده نشد.نتیجهگیری: سلولهای B این توانایی را دارند که در شرایط کشت In vitro و تحریک با محرکهای متفاوت، همانند محیط In vivo به ردهی سلولی پلاسمابلاست تمایز یابند. با این وجود، برای دستیابی به بهترین شرایط جهت تمایز سلولهای B، عواملی نظیر ماهیت تحریک، مدت زمان تحریک و استفاده از محرکهای متفاوت که نقش مهمی دارند، باید مد نظر قرار گیرند.

چکیده English
Background: Transformation and differentiation of activated B-cell to plasmacells and also memory cells depend on signaling from B-cell receptors. The signals from antigen and cytokine receptors on the surface of B cells lead to induce the expression of specific transcription factors, which finally determine the fate of B cells.Methods: Peripheral blood mononuclear cells (PBMC) were isolated via ficoll gradient and then purified B cells were separated using magnetic-activated cell sorting (MACS). Pure B cells were cultured in Roswell Park Memorial Institute 1640 (RPMI1640) culture media at the presence of purified anti-human CD40 antibody and anti-immunoglobulin M f (ab)´2 or lipopolysaccharide (LPS) and anti-human CD40 antibody that induced B-cells differentiation to plasmablasts which was assessed with 3 markers (CD38+, CD27+, IgM-) and analyzed via flow cytometry.Findings: In stimulation of B cells with purified anti-human CD40 antibody and anti-IgM f (ab)´2 or LPS through cross-linking B-cell receptor, the majority of B cells remained alive and differentiated to another lineage of B cells (plasmablast: CD38+, CD27+, IgM-). There was no significant statistical difference between expressions of plasmablast markers in two states of stimulation.Conclusion: B cells can be stimulated and differentiated to plasmablasts in vitro similar to in vivo condition. However, to achieve the best outcome in the differentiation of B cells, we should consider the nature of stimulator, the time of incubation, and the type of stimulators.
