Among these nanocomposites, poly (3-hydroxybutyrate)/nano-hydroxyapatite (PHB/nHA) has favorable porosity.Methods: Nanocomposite scaffolds of PHB/nHA were prepared via solvent-casting and particulate leaching technique. NHA constituted 0-10% of weight of polymers. The porosity of the samples was measured by diffusion method. Scanning electron microscopy (SEM) was used to evaluate the morphology of prepared scaffolds and size of nano-particles in the polymer matrix. Distribution of nHA in scaffold was investigated by energy dispersive X-ray (EDX). Degradation of scaffolds was studied by SEM, Fourier transform infrared (FTIR) spectroscopy, and weighting samples before and after degradation in phosphate-buffered saline (PBS) solution.Findings: Porosity percentage was not decreased by increasing nHA content. Size of HA particles on wall of scaffold porosity was in nano-scale and distribution of nano particles in polymer matrix was uniform. Type of PHB/nHA scaffold degradation in PBS solution was bulk degradation. Conclusion: Prepared scaffolds had good interconnectivity. Size and percentage of porosity was acceptable for cell growth, attachment, and seepage in tissue engineering. Therefore, these new PHB/nHA nanocomposite scaffolds may serve as a 3D substrate in tissue engineering.}, keywords = {Solvent-casting,Particulate leaching,Poly (3-hydroxybutyrate),Hydroxyapatit,Nanocomposite,Scaffold}, title_fa = {بررسی مورفولوژی و تخریب داربست نانوکامپوزیتی پلی(هیدروکسی‌بوتیرات)/ نانوهیدروکسی‌آپاتیت (PHB/nHA) مورد استفاده در مهندسی بافت}, abstract_fa = {روش‌ها: داربست‌های نانوکامپوزیتی پلی ‌(هیدروکسی‌بوتیرات)/نانو‌هیدروکسی‌آپتایت (PHB/nHA) به روش قالب‌گیری محلولی/ شستن ذرات تهیه شده‌اند. مقدار nHA در داربست‌ها از 0 تا 10 درصد‌ وزنی نسبت به پلیمر متغیر است. محاسبه‌ی درصد تخلخل داربست‌ها به روش نفوذ آب بدون ‌‌یون صورت گرفته است. ریخت تخلخل‌ها، پراکندگی آن‌ها، اندازه‌ی ذرات nHA و پراکندگی آن‌ها در بستر پلیمری، توسط آزمون‌های SEM ((Scanning electron microscope و EDX (Energy dispersive X-ray) مشخص شده‌اند. تخریب نمونه‌های داربستی در محیط خارج ‌بدن در Phosphate buffered saline (PBS) و دمای °C2 ± 38 به‌ مدت 102 روز انجام شده ‌است. به منظور مطالعه‌ی تخریب داربست‌ها، کاهش وزن نمونه‌ها با گذشت زمان اندازه‌گیری شده و آزمون‌های FTIR (Fourier transform infrared) و SEM قبل و پس از تخریب روی نمونه‌ها انجام گرفته است.یافته‌ها: درصد تخلخل داربست‌ها با افزایش درصد nHA کاهش نمی‌یابد. تخلخل‌ها با یکدیگر ارتباط مناسبی دارند و ابعاد آن‌ها از 200 تا 250 میکرون متغیر است. اندازه‌ی ذرات nHA در بستر پلیمری زیر 100 نانومتر می‌باشد. مطالعه‌ی تخریب نمونه‌های داربستی در محیط خارج‌ بدن در PBS و دمای °C2 ± 38 به‌ مدت 102 روز انجام شده‌ است. با بررسی تخریب داربست‌ها مشخص گردید که تخریب آن‌ها به‌صورت توده‌ای بوده است و بیشترین میزان تخریب مربوط به داربست تهیه شده با 5/7 درصد وزنی nHA می‌باشد.نتیجه‌گیری: با توجه به نتایج این پژوهش، می‌توان از داربست‌های نانوکامپوزیتی سه ‌بعدی PHB/nHA که دارای تخلخل‌هایی باز و درصد تخلخل زیاد هستند، در مهندسی بافت استخوان برای کشت و رشد سلول روی آن استفاده نمود.}, keywords_fa = {داربست,نانوکامپوزیت,پلی(هیدروکسیبوتیرات),نانوهیدروکسیآپاتیت,مهندسی بافت}, url = {https://jims.mui.ac.ir/article_13732.html}, eprint = {https://jims.mui.ac.ir/article_13732_e37b721a929960567a51ec29f69ffadd.pdf} }
