Due to phenotypic similarities of most species of Leishmania, reservoir variety, and different clinical manifestations of the disease, identification of species is necessary. In this study, the isolates which caused human CL in Isfahan region were characterized using internal transcribed spacer (ITS) polymerase chain reaction (PCR) technique. Methods: Among 60 CL suspected cases which referred to Skin Diseases and Leishmaniasis Research Center, Isfahan, Iran, the DNA isolates of patients with positive direct cutaneous microscopy were selected. Cultures were then extracted and the species of the etiologic agents of the disease were determined by ITS-PCR. Findings: Leishmania major was found as the predominant species (100%) which caused CL in Isfahan region. Conclusion: Due to phenotypic similarities among different species of Leishmania, species identification is not possible by direct microscopy and culture. Considering the endemicity of CL in Isfahan region, 3 substantial factors including reservoir combating, treatment of patients and species characterization are of importance for disease control. Keywords: Cutaneous leishmaniasis, Leishmania major.}, keywords = {}, title_fa = {تشخیص و جداسازی عامل لیشمانیوز جلدی در اصفهان با روش ITS-PCR}, abstract_fa = {مقدمه: لیشمانیوز جلدی، بیماری انگلی است که به عنوان یک مشکل بهداشتی در بخش‌هایی از ایران به ویژه در استان اصفهان محسوب می‌شود. به علت تشابهات فنوتیپی اغلب گونه‌های Leishmania و مخازن متفاوت بیماری در دو شکل خشک و مرطوب، تعیین گونه‌ی انگل لازم می‌باشد. در این مطالعه با استفاده از تکنیک PCR (Polymerase chain reaction) گونه‌های انگل عامل لیشمانیازیس مشخص شد. روش‌ها: از 60 مورد مشکوک به لیشمانیوز جلدی مراجعه کننده به مرکز تحقیقات بیماری‌های پوستی و سالک، بیمارانی که آزمایش مستقیم جلدی آن‌ها پس از رنگ‌آمیزی با گیمسا، از نظر بیماری لیشمانیوز مثبت شده بود، انتخاب شدند و پس از تهیه‌ی کشت و استخراج DNA به روش ITS-PCR (Internal transcribed spacers-polymerase chain reaction) گونه‌ی عامل بیماری تعیین گردید. یافته‌ها: در این مطالعه با استفاده از روش ITS-PCR، 60 ایزوله مورد بررسی قرار گرفت و گونه‌ی غالب L.major گزارش شد. نتیجه‌گیری: تمام گونه‌های انگل از نظر مورفولوژیکی یکسان می‌باشند و تشخیص گونه‌ها به روش‌های پارازیتولوژی (میکروسکوپی و کشت) امکان‌پذیر نیست. با توجه به این که اصفهان یکی از کانون‌های آندمیک بیماری لیشمانیوز جلدی است، بنابراین جهت مبارزه با مخازن، پیش‌گیری و درمان بیماری، شناسایی گونه‌های انگل از اهمیت بسزایی برخوردار می‌باشد. واژگان کلیدی: لیشمانیوز جلدی، لیشمانیا ماژور}, keywords_fa = {}, url = {https://jims.mui.ac.ir/article_13738.html}, eprint = {https://jims.mui.ac.ir/article_13738_39cfb6d8c339f9fd91eedb25cf4c64e4.pdf} }
