It also increased the bioavailability of pro-angiogenic factors including vascular endothelial growth factor and transforming growth factor-β. A cytosine (C) thymine (T) single nucleotide polymorphism (SNP) at position 1562 of the type IV collagenase promoter has been reported to affect gene expression. The purpose of this study was to investigate the association between the C(-1562)T polymorphism and risk of lung cancer in different age groups in Iran population. Methods: Genotyping of C(-1562)T polymorphism in type IV collagenase gene was performed by taking out genomic DNA from blood samples of 120 patients with lung cancer and 100 age-matched controls by restriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR). Chi-square test was used to calculate adjusted odds ratio (OR) and 95% confidence interval (95% CI). All analyses were performed in SPSS. Findings: Distribution of C(-1562)T genotype in type IV collagenase promoter was significantly associated with the risk of lung cancer in the age group of < 60 years (OR = 19.89; 95% CI = 3.21-120.60). Conclusion: Our results indicated that C(-1562)T polymorphism in type IV collagenase gene affects the risk of lung cancer in different age groups, i.e. aging less than 60 years was significantly related with initiation of lung cancer. Keywords: Type IV collagenase gene, Restriction fragment length polymorphism-polymerase chain reaction, Promoter polymorphism, Lung cancer}, keywords = {}, title_fa = {ارتباط پلی‌مورفیسم 1562 در ژن کلاژناز نوع IV با کاهش سن بیماری در بیماران مبتلا به سرطان ریه}, abstract_fa = {مقدمه: کلاژناز نوع IV می‌تواند کلاژن نوع IV را که جزء ساختاری اصلی غشای پایه است، تجزیه کند و سبب افزایش قابلیت دسترسی فاکتورهای پروآنژیوژنیک شامل فاکتور رشد اندوتلیال رگی و فاکتور رشد تغییر‌دهنده‌ی β ‌شود. طبق گزارش‌ها پلی‌مورفیسم تک نوکلئوتیدی سیتوزین به تیمین در موقعیت 1562 در پروموتور کلاژناز نوع IV بر بیان ژن اثر می‌گذارد. هدف از این مطالعه، جستجوی ارتباط بین پلی‌مورفیسم 1562-C/T با خطر سرطان ریه در گروه‌های سنی بود. روش‌ها: در 120 بیمار مبتلا به سرطان ریه و 100 فرد مشابه آن‌ها از لحاظ سنی به عنوان شاهد تعیین ژنوتیپ پلی‌مورفیسم 1562-C/T ژن کلاژناز IV انجام شد. این کار توسط استخراج DNA ژنومی از نمونه‌های خونی و با استفاده از تکنیک چند شکلی طولی قطعات DNA با آنزیم‌های محدودالاثر- واکنش زنجیره‌ای پلیمراز (Restriction fragment length polymorphism-Polymerase chain reaction یا RFLP-PCR) صورت گرفت. افراد مبتلا به سرطان در دو گروه سنی کمتر از 60 سال و مساوی یا بیشتر از 60 سال بررسی شدند. محاسبه‌ی خطر نسبت افزاینده (Odds ratio یا OR) با فاصله‌ی اطمینان (Confidence interval یا CI) 95 درصد توسط نرم‌افزار SPSS انجام شد. یافته‌ها: توزیع ژنوتیپ 1562-C/T در پروموتور کلاژناز نوع IV به میزان قابل ملاحظه‌ای با خطر سرطان ریه در گروه سنی کمتر از 60 سال مرتبط بود (89/19 = OR، 60/120-21/3 = CI). نتیجه‌گیری: پلی‌مورفیسم 1562- C/Tدر ژن کلاژناز نوع IV بر افزایش خطر بروز سرطان ریه در افراد کمتر از 60 سال اثر دارد. واژگان کلیدی: کلاژناز نوع IV، واکنش زنجیره‌ای پلیمراز، پروموتور، پلی‌مورفیسم، سرطان ریه}, keywords_fa = {}, url = {https://jims.mui.ac.ir/article_13871.html}, eprint = {https://jims.mui.ac.ir/article_13871_7f104940931b5aaeeccd15a5b8f3c6bf.pdf} }
