The use of bacteriocins as a substitute for currently used antibiotics is promising. The isolated nitrogen-fixing Bacillus bacteria might be a potential candidate strain as an alternative source of peptide antibiotics in the future. Methods: A total of 15 nitrogen-fixing Bacillus strains with antibacterial activity were isolated from soil samples. Bacteriocin-producing strains against the indicator strains were co-cultured and isolated. The antibacterial activity of bacteriocin was detected by spot-on-the lawn method with different bacteria. Effects of proteolytic enzymes, pH, and heat treatment on bacteriocin activity were also determined. Findings: All isolated spore forming Bacillus strains had an inhibitory effect on Listeria monocytogenes and Staphylococcus aureus. Some of them had an inhibitory effect on methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococcus (VRE). None of them had an inhibitory effect on Gram-negative bacteria. The antimicrobial compounds produced by these novel strains were inactivated by the proteolytic enzymes which demonstrated their proteinaceous nature. These bacteriocins were also active in a wide range of pH and temperature. Conclusion: In the present study, new bacteriocin-producing nitrogen-fixing Bacillus strains with anti-MRSA and anti-VRE activity were isolated from the environment and their biochemical properties were determined. Keywords: Nitrogen-fixing Bacillus strains, Bacteriocin, Methicillin-resistant Staphylococcus aureus, Vancomycin-resistant Enterococcus faecalis}, keywords = {}, title_fa = {باکتریوسین با فعالیت ضد میکروبی وسیع تولید‌شده توسط ایزوله‌ها‌ی جدید باسیلوس تثبیت‌کننده‌ی نیتروژن}, abstract_fa = {مقدمه: به تازگی شیوع دوباره‌ی عفونت‌ها‌ به دلیل پیدایش باکتری‌ها‌ی مقاوم به آنتی‌بیوتیک‌ها‌ گسترش یافته است. باکتریوسین‌ها‌ی تولید‌شده توسط باکتری‌ها‌ برای استفاده به جای آنتی‌بیوتیک‌ها‌ی مرسوم در تیمار عفونت‌ها‌ی انسانی بسیار مناسب هستند و در سال‌ها‌ی اخیر توجه زیادی را به خود جلب نموده‌اند. باکتریوسین‌ها‌ی تولید‌شده توسط باکتری‌ها‌ی متعلق به جنس باسیلوس می‌توانند نامزد بالقوه‌ای برای استفاده به عنوان عوامل ضد میکروبی جدید در آینده باشند. روش‌ها: 15 ایزوله‌ی باسیلوس تثبیت‌کننده‌ی نیتروژن دارای فعالیت ضد میکروبی از خاک جدا شدند. همچنین اثر آنزیم‌ها‌ی پروتئولیتیک و تغییرات pH و حرارت روی فعالیت باکتریوسین تولیدی بررسی گردید. یافته‌ها: تمام ایزوله‌ها‌ی باسیلوس جداشده دارای اثر مهاری بر رشد باکتری‌ها‌ی بیماری‌زای لیستریا مونوسیتوژنز و استافیلوکوکوس آرئوس بودند. برخی از ایزوله‌ها‌ی جدا‌شده باعث مهار رشد استافیلوکوکوس آرئوس مقاوم به متی‌سیلین (Methicillin-resistant staphylococcus aureus یا MRSA) و انتروکوکوس فکالیس مقاوم به وانکومایسین (Vancomycin-resistant enterococci یا VRE) شدند، ولی هیچ کدام از ایزوله‌ها‌ی جداشده اثری در مهار رشد باکتری‌ها‌ی گرم منفی نداشتند. ترکیبات ضد میکروبی تولید‌شده توسط باسیلوس‌ها‌ی تثبیت‌کننده‌ی نیتروژن به وسیله‌ی آنزیم‌ها‌ی پروتئولیتیک غیر فعال شدند ولی در برابر طیف وسیعی از تغییرات pH و حرارت فعالیت خود را حفظ نمودند. نتیجه‌گیری: در این تحقیق جداسازی باکتری‌ها‌ی باسیلوس تثبیت‌کننده‌ی نیتروژن دارای اثر مهاری بر روی سویه‌ها‌ی MRSA و VRE مورد مطالعه قرار گرفت. واژگان کلیدی: باسیلوس تثبیت‌کننده‌ی نیتروژن، باکتریوسین، استافیلوکوکوس آرئوس مقاوم به متی‌سیلین، انتروکوکوس فکالیس مقاوم به وانکومایسین}, keywords_fa = {}, url = {https://jims.mui.ac.ir/article_13973.html}, eprint = {https://jims.mui.ac.ir/article_13973_33c3406a32a8ac0fff48d7ecdcecb85e.pdf} }
