The aging process is frequently affected by environmental factors. Caloric restriction is by far the most effective and established environmental manipulation for extending lifespan in various animal models. However, the precise mechanisms by which caloric restriction affects lifespan are still not clear. Epigenetic and molecular mechanisms have recently been recognized as major contributors to nutrition-related longevity and aging control. The underlying molecular mechanisms for this effect include a lower rate of accrual of tissue oxidative damage that is associated with a significantly lower rate of mitochondrial free radical generation. Two primary epigenetic codes, DNA methylation and histone modification, are believed to dynamically influence chromatin structure and hence modify the expression of relevant genes. In this review, we assessed current advances in epigenetic regulation in response to caloric restriction and how this affects cellular senescence, aging, and potential extension of a healthy lifespan in humans. Enhanced understanding of the important role of epigenetic in the control of the aging process through caloric restriction may lead to clinical advances in the prevention and therapy of human aging-associated diseases. Keywords: Caloric restriction, Epigenetic, Aging}, keywords = {}, title_fa = {مکانیسم‌های اثر محدودیت کالری در به تعویق انداختن پیری: مروری بر شواهد موجود}, abstract_fa = { مکانیسم پیری موضوع تحقیقات زیادی می‌باشد و مداخلات بالقوه را برای به تأخیر انداختن پیری و بیماری‌های مخرب وابسته به پیری در انسان تسهیل کرده است. فرایند پیری تحت تأثیر عوامل محیطی قرار می‌گیرد. محدودیت کالری مؤثرترین و قابل دستکاری‌ترین عامل محیطی برای افزایش طول عمر در مدل‌های حیوانی مختلف می‌باشد. اگر چه مکانیسم دقیقی که محدودیت کالری روی طول عمر اثر می‌گذارد، هنوز روشن نیست؛ اما به تازگی مکانیسم‌های اپی‌ﮊنتیک و مکانیسم‌های مولکولی به عنوان عامل اصلی برای کنترل طول عمر و پیری وابسته به تغذیه شناخته شده‌اند. مکانیسم مولکولی کاهش آسیب اکسیداتیو بافتی با کاهش تولید رادیکال‌های آزاد میتوکندریایی و یا تنظیم مسیرهای متابولیکی همراه می‌باشد. در مکانیسم اپی‌ژنتیک دو کد اپی‌ﮊنتیک اصلی، متیلاسیون DNA و تغییرات هیستونی مطرح هستند و به نظر می‌رسد که به طور بالقوه بر ساختمان کروماتین و در نتیجه روی بیان ﮊنی ﮊن‌های مربوط اثر می‌گذارد. در این مقاله‌ی مروری ما پیشرفت‌های فعلی در تنظیم مولکولی و اپی‌ﮊنتیک در پاسخ به محدودیت کالری و نحوه‌ی اثر محدودیت کالری را بر پیری سلولی و افزایش توانایی و طول عمر در یک زندگی سالم در انسان‌ها بررسی کردیم.واژگان کلیدی: محدودیت کالری، اپی‌ژنتیک، پیری}, keywords_fa = {}, url = {https://jims.mui.ac.ir/article_13974.html}, eprint = {https://jims.mui.ac.ir/article_13974_673e80ab003d3a099df6b6ed704dc169.pdf} }
