Recent research has proved that these derivatives (such as allicin) are cytotoxic and therapeutic effects of garlic are in fact due to an odorless compound (S-allyl-cysteine). Considering the small number of studies in this field, we evaluated the effectiveness of S-allyl-cysteine on survival, apoptosis, and proliferation of Leishmania major promastigotes in vitro.Methods: In this study Leishmania major promastigotes (MRHO/IR/75/ER) were used. The parasites were first cultured and then treated with S-allyl-cysteine at end concentrations of 2.5, 5.0, 10.0, 20.0, 40.0, and 50.0 mM. Their survival, apoptosis, and proliferation was assessed after 72 hours.Findings: None of the concentrations of S-allyl-cysteine could induce apoptosis in parasite promastigotes. Lower concentrations (5.0 and 10.0 mM) of S-allyl-cysteine had greatest effect on proliferation. On the other hand, direct observation showed that cultures comprising S-allyl-cysteine at end concentration of 20.0, 40.0, and 50.0 mM formed more rosettes.Conclusion: The antioxidant effects of S-allyl-cysteine have been previously proved. Similarly, this study showed that some concentrations of S-allyl-cysteine could affect the survival and growth of promastigotes in culture medium. Therefore, this substance seems to be a good stimulating factor for growth of parasite promastigotes in culture media.}, keywords = {Leishmania major,S-allyl-cysteine,Apoptosis,Proliferation}, title_fa = {بررسی میزان اثربخشی ماده‌ی S-allyl-cysteine بر بقا، آپوپتوز و پرولیفراسیون پروماستیگوت‌های انگل Leishmania major در شرایط برون‌تنی}, abstract_fa = {مقدمه: تحقیقات بی‌شماری در راستای عصاره‌های روغنی سیر علیه انگل Leishmania انجام شده است، ولی مطالعات اخیر ثابت کرده‌ است که این عصاره‌ها از جمله آلیسین خود سایتوتوکسیک هستند و خاصیت درمانی سیر مربوط به ماده‌ای بی‌بو به نام SAC (S-allyl-cysteine) می‌باشد. از آن جایی که مطالعات کمی بر اثربخشی این ماده بر انگل Leishmania انجام شده است، در این مطالعه، میزان اثربخشی SAC بر بقا، آپوپتوز و پرولیفراسیون انگل Leishmania major در شرایط برون‌تنی بررسی شد.روش‌ها: در این مطالعه از سویه‌ی استاندارد Leishmania major (MRHO/IR/75/ER) استفاده شد. پس از کشت، پروماستیگوت‌ها با غلظت‌های 5/2، 5، 10، 20، 40 و 50 میلی‌مولار از SAC مواجه شدند. پس از 72 ساعت، تعداد انگل‌ها، آپوپتوز و پرولیفراسیون آن‌ها بررسی گردید.یافته‌ها: هیچ کدام از غلظت‌های SAC باعث القای آپوپتوز در انگل نشد. بررسی پرولیفراسیون مشخص کرد که غلظت‌های 5 و 10 میلی‌مولار دارای بیشترین اثر بر میزان تکثیر بودند. از طرف دیگر، مشاهده‌ی مستقیم مشخص کرد که محیط‌های کشت حاوی غلظت‌های نهایی 20، 40 و 50 میلی‌مولار دارای تعداد بیشتری اشکال روزت (Roset) می‌باشند.نتیجه‌گیری: از آن جایی که خاصیت آنتی‌اکسیدانی SAC ثابت شده است و این مطالعه نیز ثابت کرد که غلظت‌هایی از SAC می‌تواند اثر مناسبی بر بقا و افزایش رشد پروماستیگوت‌ها در محیط کشت داشته باشد، به نظر می‌رسد که این ماده به عنوان عاملی محرک جهت رشد پروماستیگوت‌های انگل می‌تواند مورد استفاده قرار گیرد.}, keywords_fa = {لیشمانیا ماژور,S آلیل- سیستیین,آپوپتوز,پرولیفراسیون}, url = {https://jims.mui.ac.ir/article_14015.html}, eprint = {https://jims.mui.ac.ir/article_14015_763ccb3e77617a48d3f5503a60af1b4d.pdf} }
