Recently, there are two methods for detection of these cells; full-thickness sections and fragmented tissues.  This study aimed to compare these to methods in detecting ganglion cells; which was not done before.Methods: In this cross-sectional study done in Alzahra hospital (Isfahan, Iran) during 2011-2012, 25 samples of colon wall in patients with detected carcinoma were studied. A 1-cm slice from each sample was fixed in formalin. Another 1-cm slice from each sample was crushed and fixed on filter paper in formalin. The samples were prepared by tissue processor machine and colored by Hematoxylin and Eosin method. Both groups were coded by second person and analyzed by a unique pathologist. The number of ganglion cells was determined and recorded.Findings: The mean ± SD of counted ganglion cells was 33.96 ± 11.80 in full-thickness sections and 25.64 ± 16.90 in fragmented tissues methods. According to independent sample t-test, the difference between two methods was statistically meaningful (P = 0.049).Conclusion: According to current study, fragmented tissues method is better than the full-thickness sections method in detection of ganglion cells for diagnosis of Hirschsprung's disease.}, keywords = {Ganglion cell,Full-thickness sections,Fragmented tissues,Hirschsprung's disease}, title_fa = {بررسی و مقایسه‌ی تعداد سلول‌های گانگلیونی در ضخامت جدار روده‌ی بزرگ انسان به دو روش برش مقطعی نمونه و خرد کردن نمونه‌ی تمام ضخامت روده‌ی بزرگ}, abstract_fa = {مقدمه: برای تشخیصی هیرشپرونگ روش‌هایی مانند بیوپسی که به مشاهده‌ی گانگلیون در نمونه می‌پردازند، ارزشمند هستند. تاکنون مطالعه‌ای بر روی هموژناسیون یا همگن‌سازی بافت جهت تشخیص هیرشپرونگ گزارش نشده است. هدف از این مطالعه مقایسه‌ی همگن‌سازی نمونه از طریق خرد کردن آن نسبت به مطالعه‌ی برش‌های متعدد از نمونه و تعیین احتمال خطا در شمارش سلولی و نیز احتمال پیدایش گانگلیون بود.روش‌ها: این مطالعه از نوع مقطعی بود که در سال 1391 انجام شد. در این مطالعه یک قطعه‌ی سالم از روده‌ی بزرگ بیماران کولکتومی‌شده با تشخیص کارسینوما، جدا شد. سپس 25 برش به طول یک سانتی‌متر تهیه شد و در فرمالین قرار گرفت. هم‌زمان 25 برش دیگر به طول یک سانتی‌متر از همان روده تهیه گردید و با تیغ بیستوری خرد شد و بر روی کاغذ صافی در فرمالین قرار گرفت. لام‌های روش برش مقطعی و لام‌های حاصل از روش خرد کردن نمونه که کدهای متفاوتی داشتند توسط پاتولوژیست از نظر تعداد سلول گانگلیونی بررسی شدند. داده‌ها توسط آزمون‌های آماری 2χ و Student-t آنالیز شد.یافته‌ها: دراین مطالعه 50 نمونه انتخاب شد و هر نمونه به دو دسته تقسیم شد. دسته‌ی اول به صورت مقطعی و دسته‌ی دوم به روش خرد کردن تحت شمارش سلولی قرار گرفتند. میانگین تعداد سلول‌های مشاهده‌شده در برش مقطعی 8/11 ± 96/33 و در روش خرد کردن 9/16 ± 64/25 عدد بود. این تفاوت از نظر آماری معنی‌دار بود (049/0 = P).  نتیجه‌گیری: با توجه به وجود تفاوت معنی‌دار بین دو روش خرد کردن و برش مقطعی، در صورتی که نمونه‌های اخذشده از روده ابتدا خرد شوند و سپس رنگ‌آمیزی گردند، امکان شناسایی سلول‌های گانگلیونی امکان‌پذیرتر خواهد بود و روش خرد کردن نسبت به روش برش مقطعی ارجحیت دارد.}, keywords_fa = {سلول گانگلیونی,برش مقطعی,خرد کردن,هیرشپرونگ}, url = {https://jims.mui.ac.ir/article_14158.html}, eprint = {https://jims.mui.ac.ir/article_14158_e7661f114fb8944b035676383d4b110a.pdf} }
