Multi-drug resistant bacteria (MDR) in different parts of the hospital including the intensive care unit (ICU), cause nosocomial infections and resistance to antibiotics made them difficult to treat. Alternative methods of treatment are one way to solve this problem. Bacteriophages are antibacterial agents that specifically attack their host. In this study, Pseudomonas specific lytic phages isolated from the clinical environment and its effect on multi-drug resistant Pseudomonas aeruginosa was investigated.Methods: In a cross sectional study in 2013, isolates of Pseudomonas aeruginosa from clinical specimens of patients admitted to the intensive care unit were identified via biochemical methods. Then, antibiotic resistance patterns were determined via standard disk diffusion method according to CLSI (Clinical and Laboratory Standards Institute) and were used as indicator hosts to screen phages from water samples.Findings: 42 strains of Pseudomonas aeruginosa wre isolated from the intensive care units. The antibiotic resistant patterns of bacterial isolates were as follows: 88% to amikacin, 90% to cefepime, 85% to ceftazidime, 90% to gentamicin, 75% to imipenem and meropenem and 95% to ciprofloxacin. Lytic bacteriophage was isolated only from hospital wastewater. The isolated bacteriophage had no effect on non- multi-drug resistant Pseudomonas aeruginosa and other Gram-negative bacteria.Conclusion: Bacterial resistance to antibiotics is increasing. New alternative methods, such as phage therapy, would open new insights in treatment of multidrug resistant bacterial infections.}, keywords = {Pseudomonas aeruginosa,Bacteriophage,Phage therapy,Multi-drug resistant}, title_fa = {بررسی تأثیر باکتریوفاژ جدا شده از نمونه‌های آب بیمارستان بر سویه‌های مقاوم به درمان (MDR) سودوموناس آئروژینوزا}, abstract_fa = {مقدمه: مقاومت عفونت‌‌های باکتریایی به آنتی‌بیوتیک‌ها به علت مصرف بیش از حد آنتی‌‌بیوتیک‌ها و مکانیسم‌های متعدد مقاومت به آن‌ها توسط باکتری‌ها رو به افزایش است. باکتری‌های مقاوم به چند دارو (MDR یا Multi-drug resistance) در بخش‌های مختلف بیمارستان از جمله بخش مراقبت‌های ویژه (ICU یا Intensive care unit)، باعث بروز عفونت‌های مهم بیمارستانی شده‌اند که درمان آن‌ها با آنتی‌بیوتیک‌ها را با مشکل مواجه ساخته است. استفاده از روش‌های جایگزین مانند استفاده از باکتریوفاژها، یکی از راه‌های حل این معضل است. باکتریوفاژها یکی از عوامل مهم ضد باکتریایی می‌باشند که به طور اختصاصی به باکتری‌های میزبان خود حمله می‌کنند و باعث انهدام آن‌ها می‌شوند. هدف از این مطالعه، جداسازی فاژهای اختصاصی سودوموناس آئروژینوزا و بررسی تأثیر آن‌ها بر روی سویه‌های مقاوم به چند دارو سودوموناس آئروژینوزا بود.روش‌ها: در یک مطالعه‌ی مقطعی در سال 1392، جدایه‌های سودوموناس آئروژینوزا از نمونه‌های کلینیکی ﺑﻴمـﺎران بستری در بخش مراقبت‌های ویژه ﺑـﻪ روش بیوشیمیایی ﺗﻌﻴﻴﻦ ﻫﻮیت شد. ﺳﭙﺲ، اﻟﮕﻮی ﻣﻘﺎوﻣﺖ آﻧﺘﻲ‌ﺑﻴﻮﺗﻴﻜﻲ آن‌ﻫﺎ با استفاده از روش ﻛﺮبی ﺑاﺋﺮ بر طبق دستورالعمل CLSI (Clinical and Laboratory Standards Institute) ﺗﻌﻴـﻴﻦ و به عنوان اندیکاتور میزبان جهت جستجوی فاژ از منابع آب استفاده گردید.یافته‌ها: 42 سویه‌ی سودوموناس آئروژینوزا از بخش مراقبت‌های ویژه به دست آمد. الگوی مقاومت آنتی‌بیوتیکی نشان داد که 88 درصد ایزوله‌ها به آمیکاسین، 90 درصد به سفی‌پیم، 85 درصد به سفتازیدیم، 90 درصد به جنتامایسین، 75 درصد به ایمی‌پنم و مروپنم و همچنین 95 درصد جدایه‌ها به سیپروفلوکسازین مقاوم بودند. در آزمایش‌های جداسازی فاژ، تنها از نمونه‌ی آب فاضلاب بیمارستان باکتریوفاژ لیتیک جداسازی شد و باکتریوفاژ جداسازی شده بر روی سودوموناس آئروژینوزاهای غیر مقاوم به چند دارو و 3 سویه‌‌ی باکتری گرم منفی دیگر تأثیری نداشت.نتیجه‌گیری: با توجه به مقاومت سودوموناس آئروژینوزا به آنتی‌بیوبیک‌های رایج استفاده از روش‌های درمانی جدید مانند فاژدرمانی افق جدیدی را در درمان عفونت‌های باکتری که به تمامی آنتی بیوتیک‌ها مقاومند باز می‌کند.}, keywords_fa = {سودوموناس آئروژینوزا,باکتریوفاژ,فاژتراپی,مقاوم به چند دارو}, url = {https://jims.mui.ac.ir/article_14466.html}, eprint = {https://jims.mui.ac.ir/article_14466_6090dd8f75b0b79639b1564e26b2b8ea.pdf} }
