This toxin targets the outer membrane of polymorphnuclear cells, monocytes and macrophages. This toxin increases the cell membrane permeability that result degradation and necrosis of leukocytes. The aim of this study was to determine the frequency of PVL-positive Staphylococcus aureus and also, to determine antibiotic resistance of the isolates.Methods: The total of 130 isolates were isolated and detected as Staphylococcus aureus during the period of 8 months in Alzahra Hospital, Isfahan, Iran. Then, polymerase chain reaction (PCR) method was used to detect PVL gene. The antibiotic susceptibility of all isolates to methicillin was determined using disk diffusion and agar screening methods.Findings: Of 130 isolates, 61 (46.92%) were methicillin-resistant and 69 (53.08%) methicillin-susceptible Staphylococcus aureus isolates (MRSA and MSSA, respectively). We found that 23.08% of isolates (30/130) were positive for PVL; of them, 11 (36.33%) were of MRSA and 19 (63.67%) were of MSSA isolates.Conclusion: Despite the existence of PVL genes in MRSA isolates, MSSA isolates can also play an important role in the dissemination of this gene. Since, PVL toxin producing strains of Staphylococcus aureus are of serious threat for health, rapid and accurate detection of gene is necessary. So, it seems that achieving a rapid and repeatable method for medical centers, will help the timely diagnosis and control of PVL-producing strains. }, keywords = {Staphylococcus aureus,Panton-Valentine leukocidin,Antibiotic Resistance}, title_fa = {شیوع ژن لکوسیدین پنتون ولنتین در سویه‌های استافیلوکوکوس اورئوس جدا شده از بیماران بیمارستان الزهرای (س) اصفهان}, abstract_fa = {مقدمه: لکوسیدین پنتون ولنتین (PVL یا Panton-Valentine leukocidin) یک توکسین گاما است که توسط استافیلوکوکوس اورئوس تولید می‌شود. این توکسین ضد غشای خارجی سلول‌های پلی‌مورفونوکلئور، مونوسیت‌ها و ماکروفاژها عمل می‌کند و باعث افزایش قدرت نفوذ پذیری غشای سلولی و در نتیجه، تخریب لکوسیت‌ها و نکروز بافت می‌شود. هدف از این مطالعه، بررسی میزان شیوع ایزوله‌های استافیلوکوکوس اورئوس PVL مثبت و تعیین الگوی مقاومت آنتی‌بیوتیکی ایزوله‌ها بود.روش‌ها: در طی هشت ماه، 130 ایزوله‌ی  استافیلوکوکوس اورئوس از نمونه‌های بالینی بیمارستان الزهرای (س) اصفهان جمع‌آوری شد. ایزوله‌های دارای ژن PVL با استفاده از روش Polymerase chain reaction (PCR) شناسایی شد. مقاومت ایزوله‌ها نسبت به متی‌سیلین و سایر آنتی‌بیوتیک‌ها به روش انتشار از دیسک بررسی شد و از روش Agar Screen برای تأیید ایزوله‌های مقاوم به متی‌سیلین استفاده شد.یافته‌ها: از مجموع 130 ایزوله‌ی استافیلوکوکوس اورئوس، 61 ایزوله (92/46 درصد) مقاوم به متی‌سیلین (MRSA یا Methicillin-resistant Staphylococcus aureus) و 69 ایزوله (08/53 درصد) حساس به متی‌سیلین (MSSA یا Methicillin-susceptible Staphylococcus aureus) بود. 30 ایزوله (08/23 درصد) از نظر وجود ژن PVI مثبت بود. از میان ایزوله‌های PVL مثبت، 19 مورد (33/63 درصد) MSSA و 11 مورد (67/36 درصد) MRSA بود.نتیجه‌گیری: با وجود تأکید بر وجود ژن PVL در ایزوله‌های MRSA، ایزوله‌های MSSA نیز می‌توانند نفش مهمی در انتشار این ژن ایفا کنند و با توجه به این که تولید توکسین PVL در سویه‌های استافیلوکوکوس اورئوس یک تهدید جدی برای سلامتی افراد محسوب می‌شود، تشخیص دقیق و سریع این ژن در باکتری فوق امری ضروری به حساب می‌آید. به نظر می‌رسد، دستیابی به یک روش سریع و تکرار پذیر در مراکز پزشکی برای کنترل عفونت با سویه‌های استافیلوکوکوس مولد PVL لازم و ضروری می‌باشد. }, keywords_fa = {استافیلوکوکوس اورئوس,لکوسیدین پنتون والنتین,مقاومت آنتی‌بیوتیکی}, url = {https://jims.mui.ac.ir/article_14514.html}, eprint = {https://jims.mui.ac.ir/article_14514_ea5ba768b2eeef936dde4462c7de50da.pdf} }
