Therefore, COX-2 inhibitors may play an effective role in cancer therapy. The goal of this study was to investigate the effect of ibuprofen and indomethacin, as anti-inflammatory drugs, on the K562 cell growth and proliferation.Methods: The K562 cells were cultured and treated with different concentrations of indomethacin and ibuprofen. Then, their antitumor effects on K562 cells was assessed via MTT assay [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] after 72 hours. DAPI staining (4',6-diamidino-2-phenylindole) and DNA electrophoresis were used for study of  apoptosis. Findings: Ibuprofen and indomethacin had antitumor effect on K562 and this effect increased by raise of time and concentration; so, the maximum of inhibitory effect was observed in higher concentration and 72 hours after the treatment. Statistical analysis indicated that both drugs inhibited the growth of K562 cells (P < 0.05).Conclusion: Based on the results, ibuprofen and indomethacin are effective agents for chronic myeloid leukemia therapy and prevention; but these antitumor properties would warrant further studies on the clinical application.}, keywords = {Nonsteroidal anti-inflammatory drug,Indomethacin,Ibuprofen,K562 cell line,Anti-cancer properties}, title_fa = {اثرات داروهای ضد التهابی ایبوپروفن و ایندومتاسین بر ردهی سلولی لوسمی میلوئیدی مزمن K562 در شرایط آزمایشگاهی}, abstract_fa = {مقدمه: در اغلب سرطان‌ها، میزان آنزیم سیکلواکسیژناز 2 (COX-2) به واسطه‌ی دخالت در سنتز پروستاگلاندین‌ها افزایش می‌یابد. بنابراین، مهار COX-2 می‌تواند نقش مؤثری در درمان سرطان ایفا کند. هدف از مطالعه‌ی حاضر، بررسی اثر داروهای ضد التهابی ایبوپروفن و ایندومتاسین بر رشد و تکثیر رده‌ی سلولی K562 بود. روش‌ها: ابتدا، سلول‌های  K562کشت داده شد. سپس، سلول‌ها با استفاده از غلظت‌های مختلف ایبوپروفن و ایندومتاسین مورد تیمار قرار گرفتند. خصوصیت ضدتوموری داروها بر سلول‌های K562 بعد از 72 ساعت، با استفاده از روش MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] در سه تکرار سنجیده شد. جهت مطالعه‌ی آپوپتوز از الکتروفورز  DNAو رنگ‌آمیزی DAPI (4',6-diamidino-2-phenylindole) استفاده گردید.یافته‌ها: ایبوپروفن و ایندومتاسین دارای خاصیت ضدتوموری بر روی سلول‌های K562 بودند و این اثر با افزایش زمان و غلظت افزایش یافت؛ به طوری که، در غلظت‌های بالاتر و 72 ساعت بعد از تیمار، شاهد بیشترین اثرات مهاری داروها بودیم. بررسی‌های آماری نشان داد که هر دو دارو به طور معنی‌داری قادر هستند، رشد رده‌ی K562 را مهار سازند (05/0 > P).نتیجه‌گیری: بر اساس یافته‌های ما، اثرات ضدتوموری ایبوپروفن و ایندومتاسین در پیش‌گیری و درمان لوسمی میلوئید مزمن مؤثر است؛ اما نیاز به بررسی‌های بیشتر بنیادی و بالینی وجود دارد.}, keywords_fa = {داروهای ضدالتهابی غیراستروئیدی,ایندومتاسین,ایبوپروفن,رده‌ی سلولی K562,خاصیت ضد توموری}, url = {https://jims.mui.ac.ir/article_14597.html}, eprint = {https://jims.mui.ac.ir/article_14597_94627b073ad5e10b82843c7a134aa38d.pdf} }
