Several virulence factors have been detected in uropathogenic Escherichia coli strains. P, S and Afa fimbria are the most important factors which are coding by pap, sfa and afa genes, respectively. This study aimed to determine the presence and prevalence of fimbrial virulence genes in Escherichia coli isolates from patients with urinary tract infection in Rafsanjan city, Iran.Methods: In this study, 145 urine samples were cultured for identification of Escherichia coli. The isolates were examined via standard biochemical tests. DNA extracts from the confirmed isolates were examined to determine papEF, afaBC and sfa / focDE genes using multiplex-polymerase chain reaction (PCR) method.Findings: Among the cultured samples, 122 Escherichia coli isolates were determined. Virulence genes genotyping of isolates showed that 38.50% of the isolates were possessed at least one of the three virulence genes. The papEF gene was the most prevalent (18.85%) and prevalences of sfa/focDE and afaBC were 2.45% and 0.81%, respectively. Twenty isolates (16.39%) contained both papEF and sfa/focDE genes.Conclusion: The results of this study in comparison with other researchers showed that papEF gene is in the high frequency, and then, afaBC and sfa/focDE fimbrial genes.}, keywords = {Escherichia coli,Fimbrial genes,Urinary tract infection}, title_fa = {ردیابی ژن‌های کد کننده‌ی فیمبریه‌های P، S و Afa در جدایه‌های اشریشیاکلی ازعفونت‌های ادراری}, abstract_fa = {مقدمه: اشریشیاکلی عامل مهمی در بروز عفونت مجاری ادراری کودکان و زنان می‌باشد. در سویه‌های اشریشیاکلی درگیر در عفونت‌های ادراری عوامل حدت متعددی شناسایی شده است که دسته‌ی مهمی از آن‌ها عوامل فیمبریه‌ای P، S و Afa می‌باشند که به ترتیب توسط ژن‌های papEF، afaBC و sfa/focDE کد می‌شوند. هدف از این مطالعه، تعیین حضور و فراوانی ژن‌های عوامل حدت فیمبریه‌ای در جدایه‌های اشریشیاکلی از بیماران مبتلا به عفونت ادراری در شهرستان رفسنجان بود.روش‌ها: در این بررسی، 145 نمونه‌ی ادراری جهت شناسایی اشریشیاکلی کشت داده شد. جدایه‌ها بر اساس آزمایش‌های بیوشیمیایی استاندارد تشخیص داده شد و DNA استخراج شده از جدایه‌ها ی تأیید شده به منظور تعیین ژن‌های papEF، afaBC و sfa/focDE به روش Multiplex-PCR (Multiplex-polymerase chain reaction) مورد بررسی قرار گرفت.یافته‌ها: در مجموع، 122 جدایه‌ی اشریشیاکلی از نمونه‌های ادراری کشت داده شده، شناسایی گردید. بررسی ژنوتیپی ژن‌های حدت نشان داد که 5/38 درصد از جدایه‌ها حداقل یکی از سه ژن حدت را دارا بودند. ژن papEF با 85/18 درصد بیشترین فراوانی و ژن‌های sfa/focDE و afaBC به ترتیب 45/2 و 81/0 درصد فراوانی داشتند. 20 جدایه (معادل 39/16 درصد) واجد هر دو ژن papEF و sfa/focDE بودند.نتیجه‌گیری: ژن papEF بیشترین درصد فراوانی را به خود اختصاص داد و ژن‌های فیمبریه‌ای afaBC و sfa/focDE در رتبه‌های بعدی قرار گرفتند.}, keywords_fa = {اشریشیاکلی,ژن فیمبریه‌ای,عفونت مجاری ادراری}, url = {https://jims.mui.ac.ir/article_14614.html}, eprint = {https://jims.mui.ac.ir/article_14614_8707696522f3e5333aa960a4b55f7e01.pdf} }
