Carbapenems resistance, due to the presence of metallo-beta-lactamase (MBL) genes encoding enzymes, is considered as a serious threat. This study aimed to detect metallo-beta-lactamase genes encoding enzymes in Pseudomonas aeruginosa strains isolated from non-hospitalized patients and to assess its antibiotic resistance.Methods: Fifty strains of Pseudomonas aeruginosa were isolated and antibiogram test was investigated with 7 different antibiotics. For 8 imipenem-resistant strains, phenotypic testing was performed via double disk synergy test (DDST) and combine disk methods. Finally, to confirm phenotypic methods, multiplex polymerase chain reaction (PCR) test was performed for detection of target genes.Findings: According to the results of the antibiogram test, 16% of the samples were resistant to imipenem and 8% to meropenem or ceftizoxime. DDST and Combine disk methods did not found any beta-lactamase positive strain. Results of phenotypic methods were confirmed via multiplex PCR and no metallo-beta-lactamase specimen was identified.Conclusion: According to the study, meropenem and ceftizoxime are good alternative medicines for imipenem. As appropriate phenotypic and molecular methods showed, the metallo-beta-lactamase genes were not prevalent in non-hospitalized patients.}, keywords = {Metallo-beta-lactamase,Multiplex polymerase chain reaction (PCR),Pseudomonas aeruginosa}, title_fa = {ردیابی ژن‌های متالوبتالاکتامازی 1blaSPM، 1blaIMP، 2blaIMP، 1blaVIM و 2blaVIM در Pseudomonas aeruginosa جداسازی شده از نمونه‌های بالینی مراکز درمانی جنوب تهران و بررسی مقاومت آنتی‌بیوتیکی}, abstract_fa = {مقدمه: Pseudomonas aeruginosa، یکی از عوامل مهم عفونت‌های بیمارستانی به ویژه در بیماران مبتلا به نقص سیستم ایمنی و کودکان است. مقاومت به کارباپنم‌ها، به دلیل وجود ژن‌های کد کننده‌ی آنزیم‌های متالوبتالاکتاماز تهدیدی جدی به شمار می‌آید. هدف از انجام این مطالعه، جداسازی ژن‌های کد کننده‌ی آنزیم‌های متالوبتالاکتاماز از سویه‌های Pseudomonas aeruginosa جدا شده از بیماران غیر بستری به روش Multiplex PCR (Multiplex polymerase chain reaction) و مقایسه با روش‌های فنوتیپی بود.روش‌ها: 50 سویه‌ی Pseudomonas aeruginosa جداسازی شد و سپس، آزمون آنتی‌بیوگرام با 7 آنتی‌بیوتیک مختلف انجام گرفت. برای 8 سویه‌ی مقاوم به ایمی‌پنم، آزمایش‌های فنوتیپی DDST (Double disk synergy test) و Combine disk انجام گرفت. در نهایت، برای تأیید روش‌های فنوتیپی انجام گرفته، Multiplex PCR برای ردیابی ژن‌های مورد نظر انجام شد.یافته‌ها: مطابق نتایج آنتی‌بیوگرام، الگوی مقاومت سویه‌های جدا شده نسبت به ایمی‌پنم 16 درصد و نسبت به سفتی‌زوکسیم و مروپنم 8 درصد بود و در روش DDST و Combine disk، هیچ سویه‌ی متالوبتالاکتاماز مثبتی یافت نشد. نتایج PCR تأیید کننده‌ی روش‌های فنوتیپی بودند و هیچ نمونه‌ی متالوبتالاکتاماز مثبتی شناسایی نشد.نتیجه‌گیری: با توجه به نتایج این بررسی، مروپنم و سفتی‌زوکسیم داروهای جایگزین مناسب برای ایمی‌پنم می‌باشند و با توجه به روش‌های فنوتیپی و مولکولی، ژن‌های متالوبتالاکتامازی مورد نظر در بیماران غیر بستری شیوع بالایی ندارند.}, keywords_fa = {متالوبتالاکتاماز,Multiplex polymerase chain reaction,Pseudomonas aeruginosa}, url = {https://jims.mui.ac.ir/article_14686.html}, eprint = {https://jims.mui.ac.ir/article_14686_7203cb39e9568b8b2187ce6ee4c3235e.pdf} }
