Genetic studies show that proteins associated with vitamin D, such as vitamin D receptor (VDR) are effective in the pathogenesis of obesity. This study aimed to investigate the relationship of FokI polymorphism in VDR gene and the body mass index (BMI), as an indicator of obesity, and other risk factors.Methods: In this case-control study, 91 patients with obesity (BMI > 30 kg/m2) and 100 healthy controls (BMI: 18.5-24.9 kg/m2) participated. The levels of fasting blood sugar (FBS), total cholesterol and triglyceride were measured and the BMI was determined. The genomic DNA was extracted from the peripheral blood of participants. VDR gene amplified using polymerase chain reaction (PCR) technique and FokI polymorphism was investigated via the restriction fragment lengths polymorphism (RFLP) method. The odds ratio was used to examine the relationship between the risk factors and the disease; 95% of confidence interval was used for these calculations.Findings: The frequency of F allele of FokI polymorphism was 95.00% and 88.46%, while the frequency of f allele was 5.00% and 11.53% in control and obese groups, respectively. The difference between F and f alleles in the control and obese groups was significant (P = 0.005). In the obese group, there was no significant relationship between the levels of fasting blood sugar, total cholesterol, and triglyceride and the genotypes. In the control group, significant relationship was observed between the level of fasting blood sugar and genotypes (P = 0.010).Conclusion: Our findings showed that protection against the obesity was conferred significantly when F allele existed. For this reason, the individuals with FF genotype in the control group had lower fasting blood sugar levels compared to the other genotypes.}, keywords = {Obesity,Vitamin D receptor,Polymorphism}, title_fa = {بررسی ارتباط پلی‌مورفیسم FokI ژن گیرنده‌ی ویتامین D (VDR) با چاقی}, abstract_fa = {مقدمه: چاقی، بزرگ‌ترین چالش بهداشت عمـومی در قـرن حاضـر است. مطالعات ژنتیک نشان می‌دهد که برخی پروتئین‌های مرتبط با عملکرد ویتامین D، مانند گیرنده‌ی ویتامین D (Vitamin D receptor یا VDR) در پاتوژنز چاقی مؤثر است. هدف از انجام مطالعه‌ی حاضر، بررسی ارتباط پلی‌‌مورفیسم FokI در ژن VDR با نمایه‌ی توده‌ی بدنی (Body mass index یا BMI) به عنوان شاخص چاقی و سایر فاکتورهای مؤثر در چاقی بود.روش‌ها: در این مطالعه‌ی مورد- شاهدی، 91 فرد مبتلا به چاقی (2kg/m 30 < BMI) و 100 فرد سالم (2kg/m 9/24-5/18 :BMI) شرکت نمودند. میزان قند خون ناشتا، کلسترول تام و تری‌گلیسرید اندازه‌گیری و نمایه‌ی توده‌ی بدنی مشخص شد. از نمونه‌های خون کامل افراد شرکت‌کننده، استخراج DNA صورت گرفت. ژن VDR با روش PCR (Polymerase chain reaction) تکثیر و سپس، پلی‌مورفیسم FokI با روش RFLP (Restriction fragment lengths polymorphism) بررسی گردید. از نسبت شانس، برای بررسی ارتباط بین فاکتورهای مورد نظر و بیماری استفاده شد و برای این محاسبات، بازه‌ی اطمینان 95% درنظر گرفته شد.یافته‌ها: فراوانی آلل F در گروه شاهد 00/95 و در گروه چاق 46/88 درصد بود. فراوانی آلل f نیز در گروه شاهد 00/5 و در گروه چاق 53/11 درصد بود. اختلاف بین آلل‌های F و f در دو گروه شاهد و چاق به صورت معنی‌داری وجود داشت (005/0 = P). در گروه چاق، بین میزان قند خون ناشتا، کلسترول تام و تری‌گلیسرید با ژنوتیپ‌ها ارتباط معنی‌داری وجود نداشت. در گروه شاهد، بین قند خون ناشتا و ژنوتیپ ارتباط معنی‌داری دیده شد (010/0 = P).نتیجه‌گیری: یافته‌های این پژوهش نشان می‌دهد که در جمعیت مورد مطالعه‌ی ما، آلل F به طور معنی‌داری نقش محافظتی در بروز چاقی دارد. به همین دلیل، افراد واجد ژنوتیپ FF در گروه شاهد، نسبت به سایر ژنوتیپ‌ها میزان قند خون ناشتای پایین‌تری داشتند.}, keywords_fa = {چاقی,گیرنده‌ی ویتامین D,پلی‌مورفیسم}, url = {https://jims.mui.ac.ir/article_14730.html}, eprint = {https://jims.mui.ac.ir/article_14730_90bd77d5b6d7945af0c393966b6a7a42.pdf} }
