Compared with human mesenchymal stem cells, murine mesenchymal stem cells have different features such as heterogeneity and slow growth rate. Several reports have shown that microRNAs are involved in many cell regulatory processes such as hypoxia. In this study, the effect of hypoxia was investigated on the expression of hypoxia related microRNA in murine mesenchymal stem cells isolated from adipose tissue (AD-MSC).Methods: AD-MSCs were cultured in two hypoxic and normoxic conditions. The expressions of mir-21 and mir-130a in the primary and immortality phase of AD-MSC were evaluated by using Real-time PCR technique. Also, the expression of MSCs surface markers were investigated by flow cytometry in the two mentioned phases.Findings: Our study showed the expression of mir-21 and mir-130a was increased in hypoxic conditions compared to normoxia. Also expressions of surface markers were different in primary and immortality phase.Conclusion: Considering that stem cells are sensitive to environmental oxygen levels, over-expression of mir-21 and mir-130a could promote the survival of MSCs exposed to hypoxia.}, keywords = {Hypoxia,Murine mesenchymal stem cells isolated from adipose tissue (AD-MSC),mir-21,mir-130a}, title_fa = {اثر محیط هیپوکسی روی بیان mir-21 و mir-130a در سلول‌های بنیادی مزانشیمی مشتق شده از بافت چربی موش در دو مرحله‌ی Primary و Immortality}, abstract_fa = {قدمه: خصوصیات منحصر به فرد سلول‌های بنیادی مزانشیمی، آن‌ها را تبدیل به ابزار قدرتمند در زمینه‌ی سلول‌درمانی و مهندسی ژنتیک کرده است. در مقایسه با سلول‌های بنیادی انسانی، سلول‌های بنیادی موشی دارای خصوصیات متفاوتی مانند هتروژن بودن و سرعت پایین رشد هستند. نشان داده شده است که microRNAها در بسیاری از فرایندهای تنظیمی سلول مانند هیپوکسی نقش دارند. در این مطالعه، اثر کمبود اکسیژن بر بیان microRNA‌های مرتبط با هیپوکسی در سلول‌های بنیادی مزانشیمی جدا شده از بافت چربی موش (Adipose-derived mesenchymal stem cells یا AD-MSC) مورد بررسی قرار گرفت.روش‌ها: سلول‌ها در شرایط هیپوکسی و طبیعی کشت داده شدند و با استفاده از تکنیک Real-time PCR (Real-time polymerase chain reaction)، میزان بیان mir-21 و mir-130a در دو مرحله‌ی Primary و Immortality مورد بررسی قرار گرفت. همچنین بیان نشانگرهای سطحی MSCها با تکنیک فلوسیتومتری مورد ارزیابی قرار گرفت.یافته‌ها: میزان بیان mir-21 و mir-130a در شرایط هیپوکسی افزایش یافت. میزان بیان این microRNA‌ها در مرحله‌ی Primary بالاتر از مرحله‌ی Immortality بود و بیشتر تحت تأثیر شرایط هیپوکسی قرار داشت. همچنین، میزان بیان نشانگرهای سطحی در سطح AD-MSC در دو فاز متفاوت بود.نتیجه‌گیری: در نهایت با توجه به این که سلول‌های بنیادی به غلظت اکسیژن حساس هستند و با در نظر گرفتن این که در شرایط In vivo سلول‌های بنیادی در شرایط کمبود اکسیژن قرار دارند، القای بیان mir-21 و mir-130a در این شرایط به بقا و حفاظت این سلول‌ها در مقابل آپوپتوز کمک می‌کند.}, keywords_fa = {هیپوکسی,mir-21,mir-130a,Adipose-derived Mesenchymal stem cells}, url = {https://jims.mui.ac.ir/article_14868.html}, eprint = {https://jims.mui.ac.ir/article_14868_1cc94fd9bb19905ec7ac10ce87b50d88.pdf} }
