In addition to existing therapy methods, using epigenetic factors specially investigation of microRNAs expression level has been known to predict prognosis, survival rate, and prevalence of a variety of cancers. In this study, the effect of resveratrol on expression level of miRNA34a and Snail gene in U87MG (ATCC® HTB-14™) cell line was investigated.Methods: In this experimental research, U87MG (ATCC® HTB-14™) cell was used in culture medium content of 1% fetal bovine serum (FBS) with penicillin and streptomycin antibiotics. Resveratrol at different concentrations was used to determine 50% inhibitory effect on cell growth in cultured cells using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT assay) method. Changes in microRNA and Snail gene expression were assessed using qualitative real-time polymerase chain reaction (qRT-PCR) method. ∆∆CT method was used to analyze gene expression data.Findings: Assessing inhibitory effect of resveratrol at different concentration for 48 hours in culture medium showed that the concentration of 43.23 µg/ml was able to inhibit growth of 50% of treated cells. Gene expression analysis indicated that expression level of microRNA34a decreased while Snail gene expression significantly increased compared to control cells.Conclusion: Resveratrol has ability to increase expression of microRNA34a but this increase did not resulted to decrease of Snail gene expression. So, it has not positive effect to control of metastasis pathway.}, keywords = {Glioblastoma,Real-time polymerase chain reaction,microRNA}, title_fa = {بررسی اثر داروی رزوراترول بر تغییر بیان MicroRNA34a در رده‌ی سلول‌های سرطانی U87MG}, abstract_fa = {مقدمه: گلیوبلاستومای چند شکل، به عنوان یکی از رایج‌ترین و تهاجمی‌ترین تومورهای سیستم عصبی مرکزی شناخته شده است. در کنار روش‌های درمانی موجود، استفاده از عوامل اپی‌ژنتیک و به ویژه بررسی تغییر بیان MicroRNAها جهت پیش‌بینی نرخ نجات و نیز گسترش انواع گوناگونی از سرطان‌ها مؤثر واقع شده است. در این مطالعه، اثر داروی رزوراترول بر تغییر بیان MicroRNA34a و Snail در رده‌ی سلولی U87MG (ATCC® HTB-14™) مورد بررسی قرار گرفت.روش‌ها: در این پژوهش آزمایشگاهی از رده‌ی سلولی U87MG در محیط کشت حاوی 10 درصد سرم جنین گاوی (FBS یا Fetal bovine serum) و آنتی‌بیوتیک‌های پنی‌سیلین و استرپتومایسین استفاده شد. از داروی رزوراترول با غلظت‌های مختلف جهت بررسی اثر غلظت مهار کنندگی 50 درصد رشد سلول‌ها با روش 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) استفاده گردید. میزان تغییرات MicroRNA و بیان ژن Snail با استفاده از روش کمیت‌سنجی در زمان واقعی (qRT-PCR یا Qualitative real-time polymerase chain reaction) انجام شد. روش ∆∆CT برای آنالیز داده‌های حاصل از بیان ژن استفاده شد.یافته‌ها: ارزیابی اثر مهار کنندگی داروی رزوراترول در غلظت‌های مختلف به مدت 48 ساعت در محیط کشت نشان داد که غلظت 23/43 میکروگرم در میلی‌لیتر قادر به مهار رشد 50 درصد از سلول‌های تیمار شده بود. آنالیز بیان ژن نیز نشان داد که میزان سطح MicroRNA34a در سلول‌های تیمار شده کاهش و بیان ژن Snail به مقدار قابل توجهی نسبت به سلول‌های شاهد افزایش یافت.نتیجه‌گیری: داروی رزوراترول توانایی افزایش بیان microRNA34a را دارد، اما سبب کاهش بیان Snail نمی‌شود و در نتیجه، اثر مثبتی بر کنترل مسیر متاستاز نخواهد داشت.}, keywords_fa = {گلیوبلاستوما,Real-time polymerase chain reaction,microRNA}, url = {https://jims.mui.ac.ir/article_15244.html}, eprint = {https://jims.mui.ac.ir/article_15244_8a76521be786f4621ed3444ec5b9bba3.pdf} }
