Previously, we produced a fusion protein consisted of catalytic and translocation domains of diphtheria toxin (DT386) fused to BR2, a cancer specific cell penetrating peptide. First steps of this study showed selective antiproliferative effects of DT386-BR2 on cancer cells but not on normal cell lines. The aim of the present study was evaluation of its in-vivo non-specific toxicity in healthy mice.Methods: The fusion protein was produced and purified through recombinant DNA technology. Intraperitoneal injections with various concentrations of DT386-BR2 were done in healthy mice for five consecutive days and they observed for 14 days after the last injection inspecting their food consumption, body weight, body temperature, and finally being dead or alive. Negative controls were injected with normal saline solution.Findings: There was not any significant effect on temperature, body weight, and viability of mice received various concentrations of DT386-BR2 (P < 0.05), and increasing the protein concentration did not show any adverse effects on mice.Conclusion: DT386-BR2 can be used for further pre-clinical studies to determine its pharmacokinetics/pharmacodynamics profiles and evaluation of its anticancer efficacy in suitable xenograft animal models.}, keywords = {Immunotoxins,Diphtheria toxin,Toxicity,Mice}, title_fa = {ارزیابی درون‌تن ایمنی فیوژن پروتئین DT386-BR2 به عنوان یک کاندیدای دارویی ضد سرطان در مدل موشی}, abstract_fa = {مقدمه: سرطان یکی از بزرگ‌ترین مشکلات مرتبط با سلامتی می‌باشد و با توجه به گسترش روزافزون مقاومت‌های دارویی و عوارض جانبی درمان‌های شیمیایی فعلی، تولید داروهای هدفمند با عوارض جانبی محدود، موضوع تحقیقات بسیاری در علوم پزشکی می‌باشد. در مطالعات قبلی، فیوژن پروتئینی مرکب از بخش کاتالیتیک و داخل غشایی سم دیفتری، به عنوان عامل کشنده و پپتید ضد میکروبی BR2 به عنوان عامل هدفمند کننده (DT386-BR2) تولید شد. مطالعات اولیه برون‌تن، اثرات ضد سرطانی انتخابی بر روی رده‌های سلولی سرطانی نشان داد. هدف از انجام مطالعه‌ی حاضر، ارزیابی درون‌تن سمیت ناشی از پروتئین DT386-BR2 بر روی موش‌های سفید سالم بود.روش‌ها: در ابتدا، فیوژن پروتئین DT386-BR2 با استفاده از تکنولوژی DNA نوترکیب در باکتری Escherichia coli تولید و با استفاده از کروماتوگرافی بر پایه‌ی ستون نیکل خالص‌سازی گردید و سپس، با غلظت‌های مختلف (15، 30، 60، 75، 150 و 300 میکروگرم/روز) در موش‌های سفید سالم به صورت داخل صفاقی و به مدت 5 روز متوالی تزریق گردید. هر گروه سه تایی از موش‌ها، از نظر وضعیت زنده ماندن، دمای بدن، میزان تغییرات وزن و میزان مصرف غذا برای دو هفته بعد از آخرین تزریق مورد ارزیابی قرار گرفتند. به گروه شاهد منفی نیز نرمال‌سالین تزریق شد.یافته‌ها: هیچ تفاوت معنی‌داری بین میزان زنده ماندن، دمای بدن و میزان تغییرات وزن موش‌های مواجه شده با فیوژن پروتئین در غلظت‌های مختلف در مقایسه با شاهد منفی وجود نداشت (05/0 < P) و با افزایش غلظت پروتئین، هیچ گونه عوارض سویی در موش‌های سالم مشاهده نگردید.نتیجه‌گیری: این فیوژن پروتئین، می‌تواند به عنوان یک کاندیدای دارویی مناسب در مطالعات درون‌تن پیش بالینی به منظور تعیین ویژگی‌های فارماکوکینتیک و فارماکودینامیک آن در مدل موشی زنوگرافت مورد ارزیابی قرار گیرد.}, keywords_fa = {ایمنوتوکسین,سم دیفتری,سمیت,ایمنی,موش}, url = {https://jims.mui.ac.ir/article_15267.html}, eprint = {https://jims.mui.ac.ir/article_15267_b086eb4383735a565732991e545e58ac.pdf} }
