There is a possibility that the amount of resistance and beta-lactam antibiotic resistance gene expression are different in Staphylococcus resistant strains. The aim of this study was to determine the expression of mecA and blaZ genes in methicillin-resistant S. aureus and the relationship between gene expression patters.Methods: In this experimental-analysis study, 120 clinical isolates of methicillin-resistant S. aureus were isolated from different clinical samples using phenotypic tests. To study the quality of the mecA and blaZ genes in resistant isolates, polymerase chain reaction (PCR) was used. In addition, the quantity of genes, SYBR-Green-1-based real-time PCR was applied. REST 2008 and SPSS software were used to analyze the data.Findings: Out of 120 isolates of methicillin-resistant S. aureus, 105 isolates (87.5%) had blaZ gene. The most diverse gene expression changes in sample type were seen in blood and urine samples. So, the expression on the wound and urine clinical samples was dramatically more than the other sites. In addition, significant relationship was observed between the blaZ and mecA gene expression and the kind of clinical samples.Conclusion: The results of this study suggested that different doses of antibiotics should be used to treat staphylococcal infections in different organs.}, keywords = {Gene Expression,Beta-lactams,Antibiotic Resistance,Methicillin-resistant,Staphylococcus aureus}, title_fa = {سنجش میزان بیان ژن‌های mecA و blaZ در سویه‌های Staphylococcus Aureus مقاوم به متی‌سیلین و تعیین ارتباط الگوی بیان ژنی آن‌ها}, abstract_fa = {مقدمه: مقاومت به بتالاکتام‌ها، از مهم‌ترین ویژگی‌های Staphylococcus aureus (S. aureus) به شمار می‌رود. این احتمال وجود دارد که میزان مقاومت در سویه‌های مقاوم و بیان ژن‌های عامل مقاومت به آنتی‌بیوتیک‌های بتالاکتامی، در S. aureus متفاوت باشد. از این رو، هدف از انجام مطالعه‌ی حاضر، تعیین میزان بیان ژن‌های mecA و blaZ در سویه‌های S. aureus مقاوم به متی‌سیلین و تعیین ارتباط الگوی بیان ژنی بود.روش‌ها: در این مطالعه‌ی تجربی- تحلیلی، 120 ایزوله‌ی بالینی S. aureus مقاوم به متی‌سیلین با آزمایش‌های فنوتیپی از نمونه‌های بالینی مختلف، جداسازی شد. جهت بررسی کیفی ژن‌های mecA و blaZ در ایزوله‌های مقاوم، از روش Polymerase chain reaction (PCR) استفاده گردید. همچنین، جهت سنجش کمی ژن‌ها، از روش Real-time PCR مبتنی بر سایبرگرین 1 استفاده شد. به منظور آنالیز داده‌های به دست آمده، از نرم‌افزارهای RG-REST و SPSS استفاده شد.یافته‌ها: در این مطالعه، از مجموع 120 ایزوله‌ی S. aureus مقاوم به متی‌سیلین، 105 ایزوله (5/87 درصد) دارای ژن blaZ بودند. در این بین، متنوع‌ترین تغییرات بیان ژنی مربوط به نمونه‌های خون و ادرار بود؛ به طوری که افزایش بیان در نمونه‌های خون و زخم و ادرار به میزان چشم‌گیری بیشتر از سایر نمونه‌های بالینی بود. همچنین، بین میزان بیان ژن mecA و blaZ در ایزوله‌های بالینی S. aureus و نمونه‌های بالینی مختلف، ارتباط معنی‌داری مشاهده شد.نتیجه‌گیری: نتایج حاصل از این پژوهش، مؤید عدم استفاده از دزهای مشابه آنتی‌بیوتیک در درمان عفونت‌های ناشی از S. aureus در بخش‌های مختلف است.}, keywords_fa = {بیان ژن,بتالاکتام,مقاومت دارویی,مقاومت به متی‌سیلین,Staphylococcus aureus}, url = {https://jims.mui.ac.ir/article_15334.html}, eprint = {https://jims.mui.ac.ir/article_15334_07ed1e90ccab1e4c6ff12f50e58748e2.pdf} }
