The clinical outcome of hepatitis B virus infection varies between individuals from spontaneous viral clearance to chronic hepatitis. This diversity may be due to variations in genes encoding cytokines. Interleukin 10, as an anti-inflammatory cytokine, may have an effect on susceptibility to hepatitis B infection. The aim of this study was to investigate the role of interleukin-10 gene promoter polymorphism (-819 C/T) (rs1800871) as a possible factor for predicting susceptibility to chronic hepatitis B virus infection.Methods: Blood samples were collected from 122 patients with hepatitis B according to positive result of the enzyme-linked immunosorbent assay (ELISA) and 138 healthy subjects as control group. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay was used for polymorphism genotyping.Findings: There was a higher frequency of CT genotype among the patients with hepatitis B (50.8%) in comparison with healthy controls (32.6%). Statistically significant differences were found in the genotype (P = 0.007) and allele (P = 0.018) frequencies of interleukin-10 gene promoter polymorphism (-819 C/T) between the patients with hepatitis B and healthy controls.Conclusion: Our findings indicate that interleukin-10 gene promoter polymorphism (-819 C/T) may be associated with chronic hepatitis B virus infection and it can serve as a possible predictive factor for the development of chronic hepatitis B infection.}, keywords = {Cytokine,Interleukin-10,Single Nucleotide Polymorphism,Chronic hepatitis B}, title_fa = {بررسی پلی‌مورفیسم پروموتر ژن اینترلوکین 10 (-819C/T) در بیماران با عفونت مزمن هپاتیت B}, abstract_fa = {مقدمه: عفونت هپاتیت B به لحاظ اپیدمیولوژیکی یک معضل عمده به حساب می‌آید. نتایج بالینی عفونت با ویروس هپاتیت B بین افراد مختلف، از پاک‌سازی خود به ‌خود ویروس از بدن تا هپاتیت مزمن، متفاوت است. این تنوع، می‌تواند به دلیل واریاسیون در ژن‌های کد کننده‌ی سیتوکاین‌ها باشد. تصور می‌شود اینترلوکین 10 (Interleukin-10)، به عنوان یک سیتوکاین ضد التهابی بر حساسیت به عفونت هپاتیت B اثرگذار باشد. این مطالعه، با هدف بررسی نقش پلی‌مورفیسم تک نوکلئوتیدی -819C/T (rs1800871) پروموتر ژن اینترلوکین 10 به عنوان نشانگر برای پیش‌آگهی از استعداد ابتلا به عفونت مزمن هپاتیت B انجام گرفت.روش‌ها: از 122 نفر که نتیجه‌ی آزمایش Enzyme-linked immunosorbent assay (ELISA) آن‌ها برای عفونت هپاتیت B مثبت شده بود (گروه مورد)، نمونه‌ی خون دریافت شد. 138 فرد سالم نیز گروه شاهد را تشکیل دادند. برای بررسی پلی‌مورفیسم و تعیین ژنوتیپ، از روش واکنش زنجیره‌ی پلیمراز- پلی‌مورفیسم طول قطعه‌ی محدود (Polymerase chain reaction-restriction fragment length polymorphism یا PCR-RFLP) استفاده شد.یافته‌ها: نتایج بیانگر فراوانی بالاتر ژنوتیپ CT در مبتلایان به هپاتیت B (8/50 درصد)، در مقایسه با افراد شاهد (6/32 درصد) بود. اختلاف معنی‌داری به لحاظ آماری در فراوانی ژنوتیپی (007/0 = P) و آللی (018/0 = P) پلی‌مورفیسم تک نوکلئوتیدی -819C/T پروموتر ژن اینترلوکین 10 بین گروه مورد (مبتلایان به هپاتیت B) و گروه شاهد مشاهده شد.نتیجه‌گیری: پلی‌مورفیسم -819C/T اینترلوکین 10 می‌تواند با عفونت مزمن هپاتیت B مرتبط باشد و این پلی‌مورفیسم را شاید بتوان به عنوان عامل احتمالی پیش‌آگهی از پیشرفت عفونت مزمن هپاتیت B در نظر گرفت.}, keywords_fa = {سیتوکاین,اینترلوکین 10,پلی‌مورفیسم تک نوکلئوتیدی,هپاتیت B مزمن}, url = {https://jims.mui.ac.ir/article_15459.html}, eprint = {https://jims.mui.ac.ir/article_15459_a1bf7d87264b686e165cfe2347cb4c41.pdf} }
