The disease is one of the most common and costly diseases in women with an estimated 176 million female infections in the world and a financial burden of about 11 billion dollars per year. In addition to existing therapies, the use of epigenetic factors, and in particular, assessment of the expression of microRNAs is effective in the diagnosis and treatment of the disease. The aim of this study was to evaluate the expression of miR-135b in endometriosis, so that through identifying changes in these microRNAs, it could be used in diagnosis and treatment.Methods: From 25 cases with endometriosis and 25 healthy subjects (control) tissue and serum samples were taken. First, RNA extraction was performed and then, using the polyA reverse transcription-polymerase chain reaction (polyA RT-PCR) and real-time polymerase chain reaction, miR-135b expression was investigated.Findings: The expression of mir-135b significantly increased in endometrial tissue of patients with endometriosis in both ectopic and uterine tissues compared to healthy women; while the expression of this microRNA in the serum samples of patients decreased compared to control group.Conclusion: Considering significant changes in both eutopic/ectopic samples and serum samples in patients with endometriosis compared with controls, miR-135b may be considered as a diagnostic and therapeutic biomarker for endometriosis.}, keywords = {Endometriosis,Serum,miR-135b}, title_fa = {miR-135b به عنوان نشانگر زیستی تشخیصی در بیماران مبتلا به اندومتریوز}, abstract_fa = {مقدمه: اندومتریوز بیماری وابسته به استروژن است که در آن، سلول‌های اندومتر که در حالت طبیعی داخل رحم وجود دارند، در خارج از رحم دیده می‌شوند. این بیماری، با شیوع تقریبی 176 میلیون زن مبتلا در جهان و بار مالی حدود 11 میلیارد دلار در سال، از شایع‌ترین و پرهزینه‌ترین بیماری‌های زنان است. در کنار روش‌های درمانی موجود، استفاده از عوامل اپی‌ژنتیک و به ویژه بررسی تغییر بیان microRNAها جهت تشخیص و درمان بیماری مؤثر واقع شده است. هدف از انجام این مطالعه، بررسی بیان miR-135b در بیماری اندومتریوز بود تا با شناسایی تغییرات این microRNAها بتوان از آن‌ها در تشخیص و درمان استفاده نمود.روش‌ها: از افراد مبتلا به اندومتریوز (25 نفر) و افراد سالم (25 نفر) نمونه‌های بافت و سرم گرفته شد. ابتدا استخراج RNA total انجام شد و سپس به روش‌های PolyA reverse transcription-polymerase chain reaction (PolyA RT-PCR) و Real time PCR بیان miR-135b مورد بررسی قرار گرفت.یافته‌ها: بیان mir-135b به میزان قابل توجهی در بافت اندومتر بیماران مبتلا به اندومتریوز در دو بافت اکتوپیک (Ectopic) و یوتوپیک (Eutopic) در مقایسه با زنان سالم، افزایش یافته بود؛ در حالی که بیان این microRNA در نمونه‌ی سرمی بیماران نسبت به گروه شاهد کاهش یافته بود.نتیجه‌گیری: با توجه به تغییرات قابل توجه بیان در افراد اندومتریوز نسبت به نمونه‌های شاهد، در نمونه‌های بافت یوتوپیک و اکتوپیک و همچنین در نمونه‌های سرمی، احتمال می‌رود بتوان miR-135b را به عنوان نشانگر زیستی تشخیصی و درمانی برای بیماری اندومتریوز به کار برد.}, keywords_fa = {اندومتریوز,سرم,miR-135b}, url = {https://jims.mui.ac.ir/article_15521.html}, eprint = {https://jims.mui.ac.ir/article_15521_134b214878a24ac1fb2e492f5ba5778b.pdf} }
