There are growing evidences that changes in level of sialic acid may cause neurodegenerative and psychotic disorders. Duo to increasing growth of these diseases, and the important role of sialic acid in causing and progression of neurogenic disease, determination of toxic levels of sialic acid is important to design the in-vitro and in-vivo experiments. So, the goal of this research was evaluation of the proper concentration of sialic acid for brain cell lines.Methods: The human astroglia cells and mice astrocytes were treated with a serial dilution of sialic acid in 96 well plates. Toxic level of sialic acid was examined using MTT assay [3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide or thiazolyl blue].Findings: The half maximal inhibitory concentration (IC50) for astroglia and astrocyte cells were 1273.3 and 3750 µM, respectively.Conclusion: Sialic acid has a dose-dependent inhibitory effect on brain cell growth with a significant growth limitation at concentrations higher than the IC50. In addition, the results showed significant differences between the two human and mouse cells, and it was suggested that instead of the mouse model, human brain cells should be used to obtain generalized results for humans.}, keywords = {Cell culture,Sialic acid,Astroglia,Astrocytes,Thiazolyl blue}, title_fa = {بررسی اثر سیالیک اسید بر رشد و بقای سلول‌های آستروسیت موشی و آستروگلیای انسانی}, abstract_fa = {مقدمه: سطح سلول‌های تمامی مهره‌داران با زنجیره‌های قندی نظیر سیالیک اسید پوشیده شده است که مقادیر زیادی از آن به صورت سیالوگلیکان در مغز مهره‌داران یافت می‌شود. شواهدی بسیاری به دست آمده است که نشان می‌دهد تغییر در میزان سیالوگلیکان، منجر به بیماری‌های نورودژنراتیو و روان‌شناختی می‌گردد. با توجه به افزایش روزافزون این دسته از بیماری‌ها و اثر تعیین کننده‌ی سیالیک اسید در فرایند ایجاد و پیشرفت بیماری‌های نوروژنیک، تعیین غلظت سمی این ماده بر سلول‌های مغزی جهت طراحی بررسی‌های درون‌تن و برون‌تن حایز اهمیت می‌باشد. از این رو، پژوهش حاضر با هدف تعیین غلظت مطلوب سالیک اسید بر رده‌ی سلول‌های مغزی انجام شد.روش‌ها: آستروگلیای انسانی و آستروسیت‌های موشی با سریال رقتی از سیالیک اسید تیمار شد و با استفاده از پلیت‌های 96 خانه‌ای و روش MTT [3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide یا تیازول آبی] حد توکسیک سیالیک اسید برای سلول‌های کشت داده شده محاسبه گردید.یافته‌ها: غلظت مهاری 50 درصد (Half maximal inhibitory concentration یا IC50) برای سلول‌های آستروگلیای انسانی و آستروسیت موشی به ترتیب 3/1273و 3750 میکرومولار به دست آمد.نتیجه‌گیری: سیالیک اسید، اثر مهاری وابسته به غلظت بر روی رشد سلول‌ها دارد؛ به طوری که در غلظت‌های بالاتر از IC50، رشد سلول‌ها محدود شد و مرگ گسترده‌ای از این سلول‌ها مشاهده گردید. همچنین، نتایج حاصل بین دو رده‌ی سلول انسان و موش اختلاف معنی‌داری نشان داد و بر این اساس، پیشنهاد می‌شود برای به دست آوردن نتایج قابل تعمیم به انسان به جای مدل موشی، از رده‌ی سلول‌های مغزی انسانی استفاده شود.}, keywords_fa = {کشت سلولی,سیالیک اسید,آستروگلیا,آستروسیت,تیازول آبی}, url = {https://jims.mui.ac.ir/article_15526.html}, eprint = {https://jims.mui.ac.ir/article_15526_633954b528ce1c6975e97fedcfc20ecb.pdf} }
