In order to study the anticancer mechanisms of this parasite, in this study the response of antisera against hydatid cyst antigens with tumor cells antigens was investigated using flow cytometry technique.Methods: Antisera against hydatid cyst antigens were prepared in rabbits, and added to the melanoma (B16F10) and breast cancer cell (4T1) lines, and also normal mouse spleen cells. The reaction was then monitored via flow cytometry.Findings: There was no significant difference in the reaction of hydatid cyst antisera (protoscolex, cell wall, and cystic fluid) with the melanoma cell line. However, the reaction of antisera against cyst fluid and cyst wall with breast cancer cells was significantly different. Additionally, the reaction of the antisera against protoscolex, wall, and cyst fluid with mouse spleen cells was less than that of normal rabbit serum (as a negative control).Conclusion: The results show that antisera against cyst wall and fluid react with the breast cancer cell line, but not with melanoma cells. These results confirm the anticancer mechanisms of this parasite. So, it may be possible to use it for selective drug delivery.}, keywords = {Flow cytometry,Malignant melanoma,Breast Cancer,Hydatid cyst}, title_fa = {بررسی واکنش آنتی‌سرم‌های ضد آنتی‌ژن‌های کیست هیداتیک با آنتی‌ژن‌های سطحی رده‌های سلولی سرطان پستان (4T1)، ملانوما (B16F10) و سلول‌های طبیعی طحال موش به روش فلوسیتومتری}, abstract_fa = {مقدمه: کیست هیداتیک، مرحله‌ی لاروی کرم Echinococcus granulosus است. اثر این انگل روی رشد سرطان در محیط کشت و در مطالعات حیوانی مختلف بررسی شده است. در جهت مطالعه‌ی مکانیسم‌‌های ضد سرطانی این انگل، در مطالعه‌ی حاضر، واکنش آنتی‌سرم‌های ضد آنتی‌ژن‌های کیست هیداتیک با آنتی‌ژن‌های سطح سلول‌های سرطانی ملانوما و پستان موش و سلول‌های طحال موش سالم به روش فلوسیتومتری مورد بررسی قرار گرفت.روش‌ها: آنتی‌سرم‌های ضد آنتی‌ژن‌های کیست هیداتیک در خرگوش تهیه شد. این آنتی‌سرم‌ها، با رده‌های سلولی ملانوما، پستان و سلول‌های طحال موش طبیعی مجاورت داده شد و واکنش آنتی‌بادی‌ها با آنتی‌ژن‌های سطحی این سلول‌ها به روش فلوسیتومتری بررسی گردید.یافته‌ها: میزان واکنش آنتی‌سرم ضد کیست هیداتیک (Protoscolex، دیواره و مایع کیست) با رده‌ی سلول‌های سرطانی ملانوما تفاوت معنی‌داری نداشت، اما میزان واکنش آنتی‌سرم‌های ضد دیواره و مایع کیست با رده‌ی سلول‌های سرطانی پستان اختلاف معنی‌داری داشت. علاوه بر این، میزان واکنش آنتی‌سرم‌های ضد Protoscolex، دیواره و مایع کیست با سلول‌های طحال موش کمتر از میزان واکنش سرم خرگوش طبیعی بود.نتیجه‌گیری: آنتی‌سرم ضد کیست هیداتیک با رده‌ی سلول‌های سرطانی ملانوما واکنش نمی‌دهد، اما با رده‌ی سلول‌های سرطانی پستان واکنش می‌دهد. این نتایج، تأیید کننده‌ی مکانیسم‌های ضد سرطان پستان این انگل می‌باشد. بنابراین، ممکن است بتوان از آن برای دارورسانی انتخابی استفاده کرد.}, keywords_fa = {فلوسیتومتری,ملانومای بدخیم,سرطان پستان,کیست هیداتیک}, url = {https://jims.mui.ac.ir/article_15685.html}, eprint = {https://jims.mui.ac.ir/article_15685_ab00f05c54b9da4b18c9b422f43a4965.pdf} }
