This drug protein is known as Anakinra, and has an effective application in the treatment of rheumatoid arthritis. This study was conducted to examine the produce of the recombinant IL-1RA protein in Escherichia coli (E. coli) strains.Methods: Codon optimization of IL-1RA gene was done using GenScript, and the gene was cloned in the pUC18 as cloning vector. Then, plasmid was cut by two restriction enzymes including NdeI and BamH1 enzymes. IL-1RA gene was purified from the agarose gel. IL-1RA gene was ligated into expression vector. The constructed expression cassette was transformed into E. coli BL21 (DE3) Origami (DE3) and Rosetta (DE3) using CaCl2 and heat shock method.Findings: Identification and confirmation of transformed colonies was performed using colony polymerase chain reaction (PCR). Induction of this gene was done with isopropyl β- d-1-thiogalactopyranoside (IPTG). The protein expression was analyzed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and western blotting techniques, and it purified by Ni nickel resin. Expression analysis of transformed E. coli strains confirmed that gene integrated into expression host. Molecular weight of expressed protein was estimated to be 16.83 kDa.Conclusion: In this study, Human IL-1RA was successfully produced in E. coli Origami with high quantity other than the rest of E. coli strains. Therefore, E. coli BL21 Origami (DE3) can be used as the suitable host for production of recombinant IL-1RA, and this technology has a potential for localization.}, keywords = {Interleukin 1 receptor antagonist protein,Recombination,Escherichia coli}, title_fa = {کلونینگ و بیان آنتاگونیست گیرنده‌ی اینترلوکین 1 نوترکیب انسانی در سیستم‌های بیانی Escherichia coli شامل (DE3) Bl21، (DE3) Origami و (DE3) Rosetta}, abstract_fa = {مقدمه: آنتاگونیست گیرنده‌ی اینترلوکین 1 (IL-1RA)، پروتئینی 153 آمینواسیدی (با وزن مولکولی 83/16 کیلودالتون) است. این پروتئین دارویی، با نام تجاری آناکینرا شناخته ‌شده است و کاربرد مؤثری در درمان آرتریت روماتوئید دارد. این مطالعه، به ‌منظور بررسی بیان پروتئین نوترکیب آنتاگونیست گیرنده‌ی اینترلوکین 1 در سویه‌های باکتری Escherichia coli (E. coli) انجام شد.روش‌ها: بهینه‌سازی ژن IL-1 RA با استفاده از GenScript انجام شد و در پلاسمید pUC18 به عنوان وکتور کلونینگ، قرار گرفت. سپس، این پلاسمید با دو آنزیم محدود کننده‌ی NdeI و BamHI برش خورد. ژن IL-1RA از روی ژل تخلیص شد. ژن IL-1RA به داخل وکتور بیانی وارد شد. سازه‌ی کاست بیانی به داخل باکتری‌های E. coli Origami، E. coli BL21 و E. coli Rosetta با روش کلرید کلسیم (CaCl2) و شوک حرارتی منتقل شد.یافته‌ها: تشخیص و تأیید کلنی‌های منتقل شده با Colony polymerase chain reaction (Colony PCR) انجام شد. القای این ژن با به کار گیری Isopropyl β- d-1-thiogalactopyranoside (IPTG) انجام گرفت. بیان پروتئین با استفاده از روش‌‌های Western blotting و Sodium dodecyl sulfate-Polyacrylamide gel electrophoresis (SDS-PAGE) تأیید شد و به وسیله‌ی رزین نیکل تخلیص گردید. آنالیز بیانی سوش‌های E. coli ترانسفورم شده تأیید کرد که ورود سازه به داخل وکتور بیانی انجام‌ شده است. وزن مولکولی پروتئین بیان‌ شده، 83/16کیلو دالتون برآورد شد.نتیجه‌گیری: در این مطالعه، آنتاگونیست گیرنده‌ی اینترلوکین 1 انسانی در باکتری E. coli Origami با مقادیر بالا نسبت به سویه‌های دیگر E. coli، به طور موفقیت‌آمیزی تولید شد. باکتری E. coli Origami می‌تواند به‌ عنوان میزبان مناسب در تولید IL-1RA نوترکیب انسانی مورد استفاده قرار گیرد و این فن‌آوری قابلیت بومی‌سازی دارد.}, keywords_fa = {آنتاگونیست گیرنده‌ی اینترلوکین 1,نوترکیبی,Escherichia coli}, url = {https://jims.mui.ac.ir/article_15926.html}, eprint = {https://jims.mui.ac.ir/article_15926_02385b7905b1c1251564e3fd06427779.pdf} }
