Epigenetic factors are involved in both of these processes. Micro RNAs (miRNAs) are epigenetic agents that accelerate the inappropriate expression of cancer. Valproic acid is a histone deacetylase inhibitor that has anticancer activity. The aim of this study was to evaluate the effect of valproic acid on miR-125b expression as one of the tumor suppressor miRNAs in breast cancer, and the expression of BCL-2 and MCL-1 anti-apoptotic genes as possible targets of miR-125b in MDA-MB-231 and MCF-7 cell lines.Methods: In this experimental study, MDA-MB-231 and MCF-7 cell lines were first cultured in optimal conditions. Cell viability was measured at different concentrations of valproic acid at 48 and 72 hours using MTT assay. The expression of miR-125b and BCL-2 and MCL-1 genes was analyzed using real-time polymerase chain reaction (PCR). The data were analyzed using one-way ANOVA test.Findings: Valproic acid significantly decreased the viability of both cell lines in dose- and time-dependent manner. It also led to a significant increase in miR-125b in both cell lines (P < 0.010). In MCF-7 cells, this drug led to a significant decrease in BCL-2 and MCL-1 genes (P < 0.001). Micro RNA (miRNA)ها عوامل اپی‌ژنتیک هستند که بیان نابه‌جای آن‌ها سرطان را شتاب می‌بخشند. والپروئیک اسید، یک داروی مهار کننده‌ی هیستون داستیلاز است که عملکرد ضد سرطانی دارد. هدف از انجام این مطالعه، بررسی اثر والپروئیک اسید بر بیان miR-125b به عنوان یکی از miRNAهای سرکوب کننده‌ی تومور در سرطان پستان و بیان ژن‌های ضد آپوپتوزی B-cell lymphoma 2 (BCL-2) و Myeloid cell leukemia-1 (MCL-1) به ‌عنوان اهداف احتمالی miR-125b در رده‌های سلولی MDA-MB-231 و Michigan Cancer Foundation-7 (MCF-7) بود.روش‌ها: در این مطالعه‌ی تجربی، ابتدا رده‌های سلولی MDA-MB-231 و MCF-7 در شرایط بهینه کشت داده شدند. زیستایی سلول‌ها در غلظت‌های متفاوت والپروئیک اسید و زمان‌های 48 و 72 ساعت با روش MTT اندازه‌گیری شد. بیان miR-125b و ژن‌های BCL-2 و MCL-1 با روش Real-time polymerase chain reaction (Real-time PCR) مورد بررسی قرار گرفتند. داده‌ها با استفاده از آزمون One-way ANOVA واکاوی شدند.یافته‌ها: والپروئیک اسید با افزایش دز و زمان، به طور معنی‌داری منجر به کاهش زیست‌پذیری هر دو رده‌ی سلولی می‌شود. همچنین، والپروئیک اسید منجر به افزایش معنی‌دار miR-125b در هر دو رده‌ی سلولی می‌گردد (010/0 > P). در سلول‌های MCF-7، این دارو منجر به کاهش معنی‌دار ژن‌های BCL-2 و MCL-1 می‌شود (001/0 > P)، اما تفاوت در بیان این دو ژن در سلول‌های تیمار شده‌ی MDA-MB-231 نسبت به گروه شاهد معنی‌دار نبود (050/0 < P).نتیجه‌گیری: احتمال می‌رود والپروئیک اسید از طریق مداخله در فرایندهای اپی‌ژنتیکی و با تأثیر بر بیان ژن‌های BCL-2 و MCL-1 می‌تواند به ‌عنوان گزینه‌ی مناسب در تحقیقات سرطان پیشنهاد شود.}, keywords_fa = {آپوپتوز,سرطان پستان,micro RNA,والپروئیک اسید}, url = {https://jims.mui.ac.ir/article_15943.html}, eprint = {https://jims.mui.ac.ir/article_15943_11280d471a1b9829515fc380cc9e4e3d.pdf} }
