اثرات تضعیفی عصاره‌ی آبی دارچین بر یادگیری و حافظه‌ی فضایی رت صحرایی نر مبتلا به دیابت شده با استرپتوزوتوسین

چکیده

مقدمه: دارچین (Cinnamonum verum) دارای اثرات درمانی همچون ضد دیابت، کاهش دهنده‌ی وزن، چربی و قند خون می‌باشد. هدف از انجام این مطالعه، بررسی اثرات عصاره‌ی آبی دارچین بر یادگیری و حافظه‌ی ‌فضایی در رت‌های مبتلا به دیابت بود.روش‌ها: در این بررسی، 28 سر رت نر بالغ از نژاد ویستار (Wistar) با محدوده‌ی وزنی 30 ± 200 گرم استفاده شد. رت‌ها به چهار گروه هفت‌تایی شاهد یا سالم، سالم دریافت کننده‌ی عصاره، مبتلا به دیابت و مبتلا به دیابت دریافت کننده‌ی عصاره تقسیم شدند. القای دیابت با تزریق داخل صفاقی 50 میلی‌گرم بر کیلوگرم استرپتوزوتوسین انجام شد. آزمایش‌ها برای سه هفته طراحی شد. در این مدت، تیمار با استفاده از عصاره با دز 200 میلی‌گرم بر کیلوگرم انجام شد. در پایان دوره، وزن رت‌های تحت تیمار با گروه شاهد مقایسه گردید. سنجش یادگیری و حافظه‌ی فضایی به کمک ماز شعاعی هشت بازویی انجام شد. داده‌ها با نرم‌افزار SPSS و با استفاده از آزمون Repeated measures ANOVA تحلیل شد.یافته‌ها: افزایش معنی‌دار (05/0 > P) در زمان سپری شدن برای یافتن غذا در رت‌های مبتلا به دیابت، دریافت کننده‌ی عصاره و مبتلا به دیابت دریافت کننده‌ی عصاره در مقایسه با گروه شاهد، نشان دهنده‌ی اثرات تضعیفی دارچین روی یادگیری و حافظه‌ی فضایی رت‌ها بود.نتیجه‌گیری: عصاره‌ی آبی دارچین، منجر به تضعیف یادگیری و حافظه‌ی فضایی رت‌های مبتلا به دیابت می‌شود. احتمال می‌رود این اثر تضعیفی در رت‌های دیابتی دریافت کننده‌ی دارچین، ناشی از افزایش قند خون و اثرات مخرب آن بر عملکرد سیستم عصبی باشد.

چکیده [English]

Background: Cinnamom (Cinnamonum verum) has therapeutic effects such as anti-diabetic, decreasing of weight and blood lipid and sugar. The aim of this study was to evaluate the effect of aqueous extract of Cinnamon on learning and spatial memory in streptozotocin-induced diabetic rats.Methods: In this study, 28 adult male Wistar rats weighting 200 ± 30 g were used. Rats were divided into four groups: control (C), control receiving extract (C+C), diabetic (D), diabetic receiving extract (D+C). Diabetes was induced by intraperitoneal injection of 50 mg/kg streptozotocin. The experiments were assigned to 3 weeks. In this time, the treatment of extract was carried out in a dose of 200 mg/kg. At the end of experiment the weight of the treated rats was compared to control groups. Evaluation of learning and spatial memory was done using radial eight-arm maze. The data were analyzed using repeated measure ANOVA by SPSS software.Findings: The significant (P < 0.05) increase of time for finding food in the diabetic, extract receiving and diabetic receiving extract rats compared to control group indicated the attenuating effects of cinnamon on rats’ learning and spatial memory.Conclusion: The aqueous extract of cinnamon attenuates the learning and the spatial memory of diabetic rats, probably due to increase of blood sugar and its destructive effects on nervous system function.
