بررسی پایداری کیت ELISA آنتی‌ژن B نوترکیب اکینوکوکوس گرانولوزوس با استفاده از روش‌های فیزیکی و باکتریواستاتیک

چکیده

مقدمه: کیست هیداتید، عامل هیداتیدوز، یک بیماری زئونوز است که توسط مرحله‌ی لاروی گونه‌های مختلف سستود اکینوکوک، اغلب به صورت کیسه‌های پر از مایع، در بافت‌های مختلف بدن ایجاد می‌شود. تشخیص این بیماری در انسان به روش‌های گوناگونی انجام می‌گردد که یکی از رایج‌ترین آن‌ها‌ روش ELISA می‌باشد. یکی از ترکیبات اصلی آنتی‌ژنیک مایع کیست هیداتیک اکینوکوکوس گرانولوزوس، آنتی‌ژن B است.روش‌ها: برای تهیه‌ی کیت، ابتدا ژن کدکننده‌ی آنتی‌ژن B در وکتور بیانی کلون و سپس، بیان و خالص گردید و در انتها، به عنوان آنتی‌ژن اصلی در چاهک‌های ELISA پوشش داده شد. در این مطالعه، با استفاده از روش‌های استاندارد، پایداری فیزیکی و باکتریواستاتیک کیت ELISA آنتی‌ژن نوترکیب مورد بررسی قرار گرفت. سنجش پایداری فیزیکی با استفاده از تست پایداری تسریع شده (نسبت Arrhenius) صورت گرفت. برای انجام، تعداد مشخصی نمونه‌ی مثبت و منفی، ابتدا با کیت مورد نظر سنجش شد؛ سپس، کیت در شرایط دمایی خاص و زمان معین قرار گرفت و بار دیگر، نمونه‌های قبلی با این کیت مورد سنجش قرار گرفت و درصد پایداری آن‌ها مشخص شد. سنجش پایداری باکتریواستاتیک، با استفاده از مواد نگه‌دارنده مانند سدیم آزاید و پروکلین 300، در غلظت‌های متفاوت با استفاده از باکتری باسیلوس سوبتیلیس صورت گرفت. برای انجام این مرحله، ابتدا غلظت‌های متفاوت از باکتری و درصدهای متفاوت از مواد نگه‌دارنده را تهیه شد؛ سپس، به محلول‌های داخل کیت اضافه گردید و اثر ضدمیکروبی آن‌ها بررسی شد تا کمترین درصد نگه‌دارنده که بیشترین خاصیت باکتریواستاتیکی را دارد، مشخص گردید.یافته‌ها: بر اساس تست‌های پایداری فیزیکی، کیت طراحی شده دارای پایداری یک ساله و عدم پایداری دو ساله در دمای 8-2 درجه‌ی سانتی‌گراد (یخچال) بود. بر اساس سنجش پایداری باکتریواستاتیک، سدیم آزاید اثر تخریبی بر روی محلول‌های داخل کیت داشت ولی، پروکلین 300 با غلظت 05/0 درصد کمترین اثر تخربی را دارا بود.نتیجه‌گیری: مطالعات پایداری فیزیکی و باکتریواستاتیک انجام شده نشان داد که کیت طراحی شده با آنتی‌ژن B نوترکیب و حاوی نگه‌دارنده‌ی پروکلین 300 با غلظت 05/0 درصد، برای مدت زمان یک سال در دمای 8-2 سانتی‌گراد پایدار است.

چکیده [English]

Background: Cystic hydatidosis disease (CHD) is a common disease of human and animal caused by contamination with the metacestode stage of echinococcus granulosus. The disease causes cyst in tissue body of patients. Antigen B is one of the important antigens in liquid cyst.Methods: Expressed and purified recombinant antigen of echinococcus granulosus was used as antigen in ELISA (Enzyme-linked immunosorbent assay) method. The handmade kit was studied with physical and bacteriostatical methods. Physical stability was assessed with accelerated stability test (Arrhenius equation). First, numeral negative and positive samples were evaluated; then, the kit was exposed in special temperatures and times; afterwards, the samples were evaluated again and compared with the first evaluation. Proclin300, Soduim azide and Bacillus subtilis were chosen in bacteriostatic method. Different concentration of bacillus and different percents of preservatives were added to solutions of the kit. Growth of bacteria was evaluated and the least percent of preservatives that had the most effect of bacteriostatic was determinated.Findings: According to stability test, the handmade kit had one-year stability and do not have stability for two years in the 2-8ºC tempreture. Sodium azide had destructive effect on solutions of the kit; but, proclin300 with the concentration of 0.05% was appropriate for solutions.Conclusion: Stability and bacteriostatic methods showed that the handmade kit can be maintained in 2-8ºC with significant activity.
