بررسی اثر درمانی داروی کلشی‌سین در آسیب دیررس ریوی ناشی از گاز خردل در مدل حیوانی

چکیده

مقدمه: در میان عوارض دیررس مصدومین گاز خردل (SM)، مشکلات ریوی مانند برونشیت مزمن و فیبروز ریوی مهم‌ترین علت ناتوانی بلندمدت در بیماران می‌باشد. کلشی‌سین دارای اثرات ضد التهابی بوده و در فیبروز ریوی ناشی از سایر علل کاربرد داشته است. بر این اساس و با توجه به نقش استرس اکسیداتیو در پاتوژنز برونشیت مزمن و فیبروز ریوی، در این مطالعه به بررسی اثر درمانی کلشی‌سین در عوارض دیررس SM و نقش استرس اکسیداتیو در مکانیسم اثر آن در مدل حیوانی پرداختیم.روش‌ها: یک ماه پس از مواجهه‌ی موش‌های سوری نر با SM، یک گروه شش تایی به عنوان گروه مداخله و یک گروه 6 تایی به عنوان شاهد مثبت انتخاب شد. پس از Sacrifice و نمونه‌گیری از موش‌ها، شاخص‌های التهاب و فیبروز ریوی و استرس اکسیداتیو در گروه مداخله با گروه شاهد مثبت مقایسه شد.یافته‌ها: نتایج، نشان‌دهنده‌ی کاهش درصد فضای آلوئولی ریه و ویتامین C و کاتالاز سرمی و افزایش ارتشاح لنفوسیت‌ها و هیدروکسی پرولین بافتی و افزایش مقدار H2O2 سرمی در گروه شاهد مثبت در مقایسه با شاهد منفی بود. در گروه مداخله افزایش ویتامین C و کاتالاز سرمی و کاهش هیدروکسی پرولین بافتی و H2O2 سرمی نسبت به گروه شاهد مثبت مشاهده گردید. علاوه بر این، در سایر موارد تفاوت معنی‌داری به دست نیامد.نتیجه‌گیری: کلشی‌سین به واسطه‌ی کاهش التهاب و استرس اکسیداتیو ناشی از گاز خردل می‌تواند در کاهش عوارض دیررس ریوی گاز خردل مؤثر باشد.

Background: Among late complications of sulfur mustard gas, respiratory problems, especially chronic bronchitis and pulmonary fibrosis, are the greatest causes of long-term disability. Colchicine has anti-inflammatory effects and also indication of its usage in pulmonary fibrosis. According to these and note to the effect of oxidative stress in pathogenesis of chronic bronchitis and pulmonary fibrosis, the therapeutic effects of Cochicine and the role of oxidative stress in its mechanism was studied.Methods: One group of mice that had inhaled sulfur mustard one month ego was treated by colchicine. After sacrificing and sampling, biochemical indeces of chronic inflammation and pulmonary fibrosis of experimental group was compared with positive and negative control groups.Finding: A significant decrease was seen in alveolar space percentage, serum vitamin C, and catalase and an increase was seen in hydroxylproline concentration, lymphocyte infiltration, and serum H2O2 in positive control group in comparison with negative control. Vitamin C and catalase increase and hydroxylproline concentration, lymphocyte infiltration and serum H2O2 decrease in experimental group in comparison with positive control group were obtained.Conclusion: Colchicine has therapeutic effects on delayed lung complications of sulfur mustard gas injury and diminishing the oxidative stress can be one of mechanisms of this drugs.
