چکیده
مقدمه: تحرک در هلیکوباکتر پیلوری یکی از مهم‌ترین فاکتورهای کلونیزاسیون است که توسط فلاژل انجام می‌شود. فلاژل از 3 قسمت پایه، قلاب و رشته تشکیل شده است. رشته دارای دو پروتئین FlaA، FlaB است که برای حرکت ضروری می‌باشند. ژن‌های کد‌کننده‌ی این دو پروتئین در سویه‌های استاندارد هلیکوباکتر پیلوری حفظ شده‌اند. هدف این مطالعه، بررسی فراوانی ژن flaA در سویه‌‌های کلینیکی هلیکوباکتر پیلوری جداشده از بیماران مراجعه‌کننده به بخش آندوسکوپی و تعیین میزان حساسیت و ویژگی آن برای شناسایی هلیکوباکتر پیلوری بود.روش‌ها: در این مطالعه 100 بیوپسی‌ معده که از مراکز درمانی تهیه‌ شده بود، کشت داده شد. بعد از تأیید وجود باکتری، DNA آن استخراج گردید و برای تعیین ژن flaA واکنش زنجیره‌ای پلیمراز (Polymerase chain reaction یا PCR) با پرایمر طراحی‌ شده انجام شد.یافته‌ها: نتیجه‌ی PCR برای تمام نمونه‌های کلینیکی مثبت بود و تمام سویه‌های کلینیکی مورد آزمایش دارای ژن flaA بودند.نتیجه‌گیری: در این مطالعه همه‌ی نمونه‌های کلینیکی دارای ژن flaA بودند. از آن جایی که این ژن حفظ شده است و دارای شباهت زیاد با سویه‌های استاندارد هلیکوباکتر پیلوری می‌باشد، بنابراین از این ژن می‌توان برای تشخیص هلیکوباکتر پیلوری استفاده کرد.

چکیده English

Background: This study aimed to survey the presence of flagellin gene flaA in isolated Helicobacter pylori.Methods: One hundred gastric biopsies were collected by a gastroenterologist and inoculated in enrichment medium under microaerophilic conditions. DNA was extracted and polymerase chain reaction (PCR) was performed using the designed primer.Findings: PCR products showed that all extracted DNA were positive for flaA gene.Conclusion: We found flaA gene to be present in all clinical isolates. As it is highly similar to standard Helicobacter pylori strains, it can be used to detect the mentioned bacteria.
