ارایه‌ی یک روش جدید و کارامد در تشخیص اتوماتیک سلول‌های لوسمی حاد لنفوئیدی (ALL) با استفاده از تلفیق شاخص‌های آشوب‌گونه، هندسی و آماری

چکیده

مقدمه: لوسمی حاد لنفوئیدی (Acute lymphocytic leukemia یا ALL) یکی از شایع‌ترین انواع سرطان خون در میان کودکان می‌باشد. با توجه به تعداد زیاد آزمایشگاه‌های تشخیص طبی، به نظر می‌رسد در آزمایشگاه‌هایی که فرد خبره‌ای به منظور تشخیص نهایی ALL حضور ندارد، نرم‌افزار می‌تواند ابزار مناسبی در جهت اهداف کمک تشخیصی باشد. هدف از انجام پژوهش حاضر، استفاده از خصوصیات هندسی و آماری هسته‌ها در ایجاد یک سیستم تشخیص اتوماتیک دقیق بود.روش‌ها: در این مطالعه با استفاده از الگوریتم تقطیع اتوماتیک، هسته‌ی سلول‌های سرطانی از تصاویر موجود جدا شد. با توجه به این که ویژگی‌های آشوب‌گونه باعث تمایز معنی‌داری در الگوهای رشته‌ای و لبه‌ها می‌گردد، در مرحله‌ی بعد سه دسته ویژگی هندسی، آماری و آشوب‌گونه از سلول‌ها استخراج گردید. به منظور تشخیص و طبقه‌بندی، الگوریتم ماشین بردار پشتیبان (Support vector machines یا SVMs) برای طبقه‌بندی مورد استفاده قرار گرفت.یافته‌ها: روش پیشنهادی بر روی 312 تصویر میکروسکوپی شامل سلول‌های سالم و سرطانی مورد ارزیابی قرار گرفت. یافته‌ها، صحت بالای ۹۲ درصد و دقت بالای ۹۳ درصد در شش گروه سلول را نشان داد. بررسی سطح زیر منحنی (Receiver operating characteristic یا ROC) نشان دهنده‌ی کارایی بالای ۹۱ درصد در روش پیشنهادی بود.نتیجه‌گیری: نتایج نشان دهنده‌ی کارامد بودن ویژگی‌های مذکور در طبقه‌بندی سلول‌های ALL است. از جمله امتیازات استفاده از ویژگی‌های آشوب‌گونه می‌توان به ایجاد تمایز در هسته‌ی سلول‌های سالم و سرطانی (که در اندازه و کروماتین تفاوت دارند) و استفاده از ویژگی‌های شکل ناهمگون سیتوپلاسم سلول‌های سالم اشاره نمود.

Background: Acute lymphoblastic leukemia (ALL) is one of the most common types of leukemia among children. Due to the large number of clinical laboratories, in those with no expert pathologist for diagnosis of leukemia, software can be a useful tool for diagnostic purposes. The aim of this study was to create an automatic detector to help diagnosis process.Methods: Using automatic segmentation algorithm, the nucleus of blast and lymphocyte cells were separated from existing images. As the chaotic characteristic caused significant difference in edges and string patterns, three geometrical, statistical, and chaotic features were derived from cells. In order to diagnosis and classification, support vector machine algorithm was used and the accuracy of classification was investigated using receiver characteristic operating curves (ROC).Findings: This study was conducted on 312 microscopic images including blast and lymphocyte cells. There was a specificity of more than 92% and an accuracy of more than 93% in six cell groups. In addition, checking out the area under the ROC curve represented more than 91% efficiency for suggested method.Conclusion: The findings indicate the effectiveness of these features in classification. Differentiation of blast and lymphocyte cells, that are different only in size of chromatin, and also uneven shape of lymphocyte cytoplasm, are of the advantages of using chaotic features.
