بررسی تأثیر ضد التهابی سیاه‏دانه بر روی موش‏های صحرایی ماده‌ی مبتلا به آنسفالومیلیت آلرژیک تجربی

چکیده

مقدمه: Multiple sclerosis (MS)، بیماری مزمن و التهابی سیستم عصبی است و پیشرفت بیماری با افزایش سیتوکاین‏های خاص ارتباط دارد. آنسفالومیلیت آلرژیک تجربی، به عنوان یک مدل حیوانی برای بیماری MS در نظر گرفته می‏شود و از آن برای ارزیابی روند بیماری و فرایند درمانی استفاده می‏گردد. با توجه به مطالعات انجام شده در زمینه‌ی فواید گیاه سیاه‌دانه، این مطالعه، با هدف بررسی تأثیر ضد التهابی سیاه‌دانه بر روی موش‏های صحرایی ماده‌ی مبتلا به آنسفالومیلیت آلرژیک تجربی انجام شد.روش‌ها: در این مطالعه، تعداد 24 سر موش صحرایی ماده‌ی بالغ نژاد Lewis (با میانگین سن 8-6 هفته و وزن 220-200 گرم) در چهار گروه (6 = n) تقسیم شدند. گروه شاهد نرمال‌سالین و موش‌های صحرایی مبتلا به آنسفالومیلیت آلرژیک تجربی (Experimental autoimmune encephalomyelitis یا EAE) تحت درمان، 05/0 سی‌سی عصاره‌ی هیدروالکلی سیاه‌دانه را در دزهای (50، 100 و 200 میلی‌گرم/کیلوگرم وزن بدن) به صورت یک روز در میان به صورت تزریق داخل صفاقی دریافت کردند. عامل نکروز دهنده‌ی تومور آلفا (Tumor necrosis factor-α یا TNF-α)، پروتئین واکنشگر C (C-Reactive protein یا CRP)، نیترات و تغییرات وزن بدن روزانه ثبت گردید. بعد از پایان دوره‌ی 15 روزه، میزان تولید TNF-α و CRP با استفاده از روش Enzyme-linked immunosorbent assay (ELISA) و میزان نیترات توسط دستگاه High-performance liquid chromatography (HPLC) اندازه‌گیری شد. اطلاعات به دست آمده، با استفاده از آزمون‌های آماری One-way ANOVA، Mann-Whitney و آزمون Duncan مورد بررسی قرار گرفت.یافته‌ها: سطوح سرمی TNF-α، CRP و نیترات سرم وابسته به دز در گروه‌های تیمار نسبت به گروه شاهد به صورت معنی‌داری کاهش نشان داد (001/0 > P).نتیجه‌گیری: به دلیل این که وضعیت التهاب در روند بیماری MS نقش به‌سزایی دارد، اثرات ضد التهابی، آنتی‏اکسیدانی و محافظتی عصاره‌ی سیاه‌دانه در دز (200 میلی‌گرم/کیلوگرم وزن بدن) موجب کاهش عوامل التهابی و بهبود وضعیت التهابی در موش‏های صحرایی مبتلا به آنسفالومیلیت آلرژیک تجربی می‏گردد.

Investigating the Anti-inflammatory Effect of Nigella Sativa in Female Rats with Experimental Allergic Encephalomyelitis

Background: Multiple sclerosis (MS) is a chronic and inflammatory disease of the nervous system, and the progression of the disease is associated with an increase in specific cytokines. Experimental allergic encephalomyelitis is considered as an animal model for MS, and is used to assess the disease process and therapeutic process. According to studies on the benefits of Nigella sativa, the aim of this study was to evaluate the anti-inflammatory effect of Nigella sativa on female rats with experimental allergic encephalomyelitis.Methods: In this study, 24 adult Lewis female rat (average age of 6 to 8 weeks, weighing 200-220 g) were selected in 4 groups of 6. The control mice received normal saline and the ones with experimental allergic encephalomyelitis received 0/05 cc of hydroalcoholic extract of Nigella sativa at doses of 50, 100, and 200 mg/kg body weight every other day as intraperitoneal injection. Symptoms of disease and body weight changes were recorded in daily bases. Tumor necrosis factor-α (TNF-α), C-reactive protein (CRP), nitrate, and body weight changes were recorded daily. After the end of the period (15 days), the production of TNF-α and CRP was measured using the enzyme-linked immunosorbent assay (ELISA) and nitrate levels using high-performance liquid chromatography (HPLC). The collected data were analyzed using one-way ANOVA and Duncan tests.Findings: Serum levels of TNF-α, CRP, and nitrate were significantly lower in treatment groups than the control group (P < 0.001).Conclusion: Because inflammation plays an important role in the process of MS, the anti-inflammatory, antioxidant, and protective effects of the extract of dandelion (200 mg/kg body weight) reduce inflammatory factors and improve inflammatory status in rats with experimental allergic encephalomyelitis.
