بررسی اثر عصاره‌ی هیدروالکلی گیاه اسطوخودوس بر روی نورون‌های پورکنژ تحت استرس شنای اجباری در موش‌های صحرایی نر با روش هیستولوژی و ایمونوهیستوشیمی

چکیده

مقدمه: بر اساس جستجوهای انجام شده، در مورد تأثیر عصاره‌ی گیاه اسطوخودوس (Lavender) بر روی نورون‌های پورکنژ تحت استرس شنای اجباری مطالعه‌ای یافت نشد. از این رو، مطالعه‌ی حاضر با هدف بررسی تأثیر عصاره‌ی گیاه اسطوخودوس بر روی نورون‌های پورکنژ تحت استرس شنای اجباری انجام شد.روش‌ها: در این مطالعه‌ی تجربی، 50 سر موش صحرایی به صورت تصادفی به 5 گروه 10تایی تقسیم شدند (10 = n). چهار گروه تحت استرس شنای اجباری به ترتیب عبارت از گروه اول دریافت کننده‌ی نرمال‌سالین، گروه دوم، سوم و چهارم مورد عصاره‌ی اسطوخودوس با دزهای 200، 400 و 600 میلی‌گرم/کیلوگرم به صورت گاواژ به مدت 14 روز دریا فت کردند. گروه پنجم، گروه شاهد (Intact) بود. پس از دوره‌ی درمان با بیهوشی عمیق، مخچه خارج و در فرمالین ثابت شد. با روش هیستولوژی و ایمونوهیستوشیمی، بررسی ریخت‌شناسی نورون‌ها انجام شد. داده‌ها با آزمون‌های Duncan و Scheff تجزیه و تحلیل شدند.یافته‌ها: میانگین تعداد نورون‌های سالم پورکنژ در حیواناتی که 400 میلی‌گرم/کیلوگرم عصاره‌ی اسطوخودوس می‌گرفتند، در مقایسه با سایر گروه‌های مورد که دزهای 200 و 600 میلی‌گرم/کیلوگرم از این عصاره و یا نرمال‌سالین دریافت می‌کردند، افزایش معنی‌داری نشان داد (001/0 > P). شکل غیر طبیعی نورون‌های پورکنژ، متراکم شدن هسته، مشخص نبودن محدوده‌ی هسته از سیتوپلاسم، افزایش اسیدوفیلی و مثبت شدن نورون‌های پورکنژ با Caspase-3 در گروه تحت استرس شنای اجباری دریافت کننده‌ی نرمال‌سالین در مقایسه با سایر گروه‌ها بیشتر بود.نتیجه‌گیری: مطالعه‌ی حاضر نشان داد که عصاره‌ی اسطوخودوس در دز 400 میلی‌گرم/کیلوگرم می‌تواند یک نقش محافظتی بر روی نورون‌های پورکنژ قشر مخچه‌ی موش‌های صحرایی تحت استرس شنای اجباری داشته باشد.

چکیده [English]

Background: There was no report about the effect of Lavandula angustifolia (lavender) on Purkinje neurons in cerebellum. The purpose of this study was to determine the effects of lavender extract on structure of Purkinje neurons during forced swimming stress.Methods: Fifty rats were randomly divided into five groups of ten. Four groups were enforced under swimming stress; the first group received gavage normal saline, the second, third, and fourth group received 200, 400, and 600 mg/kg of gavage lavender extract, respectively. The fifth group considered as intact control. The treatment period was 14 days. Rats were deeply anesthetized and sacrificed, and the cerebella dissected and fixed in formalin. Histological passage was performed for each section, and stained thereafter. The number of normal Purkinje neurons were then counted. The morphological changes in the Purkinje neurons were histologically and immunohistochemically determined. Data were statistically analyzed using Duncan and Scheffe tests.Findings: There were significant increase in mean number of normal Purkinje neurons in rats received 400 mg/kg lavender extract compared to other groups (P < 0.001). Compared to other groups, the morphological changes such as density of the nucleus in Purkinje neurons, raised cytoplasm acidophilic, and caspase-3-positive neuronal Purkinje increased in the group with swimming stress and normal saline.Conclusion: The results showed that lavender extract at 400 mg/kg dose may protect Purkinje neurons in male rats under forced swimming stress.
