مقایسه‌ی اثر آنتی‌اکسیدانی نانوذرات پلی‌لاکتیک کوگلیکولیک اسید (PLGA) حاوی عصاره‌ی جینسینگ قرمز کره‌ای (KRGE) نسبت به عصاره‌ی جینسینگ قرمز کره‌ای در درمان سندرم تخمدان پلی‌کیستیک ناشی از تستوسترون

چکیده

مقدمه: پژوهش حاضر با هدف ارزیابی و مقایسه‌ی اثرات عصاره‌ی جینسینگ قرمز کره‌ای (Korean red ginseng extract یا KRGE) و نانوذرات پلی‌لاکتیک کوگلیکولیک اسید [Poly(lactic-co-glycolic acid) یا PLGA] حاوی KRGE در درمان موش‌‌های مبتلا به سندرم تخمدان پلی‌کیستیک (Polycystic ovary syndrome یا PCOS) ناشی از تستوسترون انجام شد.روش‌ها: موش‌های ماده‌ی Naval Medical Research Institute (NMRI) پس از بلوغ به سه گروه شاهد منفی (10 سر موش سالم)، شاهد مثبت (10 سر موش تحت درمان با دارونما) و بیمار (60 سر موش) تقسیم شدند. بیماری PCOS از طریق تزریق زیرجلدی 1 میلی‌گرم در 100 میلی‌گرم در روز تستوسترون انانتات به پشت گردن موش‌ها به مدت 4 هفته در دو گروه شاهد مثبت و بیمار القا گردید. سپس گروه بیمار به صورت داخل صفاقی با غلظت‌های مختلف عصاره‌ی جینسینگ (50، 100 و 200 میکروگرم در میلی‌لیتر) و نانوذرات PLGA حاوی عصاره‌ی جینسینگ (3، 6 و 10 میکروگرم در میلی‌لیتر) برای 15 روز متوالی تحت درمان قرار گرفتند. بافت‌های تخمدان به منظور تعیین مالون دی‌آلدئید (Malondialdehyde یا MDA)، گونه‌ی فعال اکسیداتیو، گلوتاتیون احیا، ظرفیت آنتی‌اکسیدانی کل (Total Antioxidant capacity یا TAC)، گلوتاتیون S-ترانسفراز (Glutathione S-transferase یا GST) و Real-time polymerase chain reaction (RT-PCR) [برای بررسی بیان دو ژن گلوتاتیون ردوکتاز (Glutathione reductase یا GR)] و فاکتور هسته‌ای اریتروئید 2 مرتبط با فاکتور 2 (Nuclear factor erythroid 2 یا NRF-2) با جراحی برداشته شد.یافته‌ها: عصاره و نانوذرات حاوی جینسینگ، منجر به کاهش MDA و گونه‌ی فعال اکسیداتیو و همچنین، افزایش سطح گلوتاتیون احیا، TAC و فعالیت GST در موش‌های مبتلا به PCOS شد. با توجه به نتایج RT-PCR، مقادیر بیان دو ژن NRF-2 و GR در موش‌های تحت درمان افزایش یافت. بر اساس یافته‌ها، نانوذرات PLGA حاوی جینسینگ مؤثرتر از عصاره بود.نتیجه‌گیری: نانوذرات PLGA حاوی جینسینگ، در کاهش عوارض ناشی از بیماری PCOS مؤثرتر می‌باشد.

Background: This study aimed to evaluate and compare the effects of Korean red ginseng extract (KRGE) and KRGE-loaded poly(lactic-co-glycolic acid) nanoparticles (KRGE-loaded PLGA NPs) on testosterone-induced polycystic ovarian syndrome (PCOS) mice.Methods: Female Naval Medical Research Institute (NMRI) post-pubertal mice were divided into control (n = 10), sham (n = 10), and PCOS (n = 60) groups. PCOS was induced in sham and PCOS groups by subcutaneous injection of testosterone enanthate 1 mg/100mg/day in the back of the mice neck for 4 weeks; then, the PCOS group was intraperitoneally treated with different concentrations of KRGE and KRGE-loaded PLGA NPs for the consecutive 15 days. The ovarian tissues were surgically removed for determinations of malondialdehyde (MDA), reactive oxygen species, glutathione (GSH), total antioxidant capacity (TAC), glutathione S-transferase (GST), and qualitative real-time polymerase chain reaction (qRT-PCR) analysis to detect glutathione reductase (GR) and nuclear factor erythroid 2 (NRF-2) genes.Findings: Both KRGE and KRGE-loaded PLGA NPs were beneficial to decrease MDA, and oxidative agents as well as increase the level of GSH, TAC, and GST in PCOS mice. According to qRT-PCR analysis, the expressions of NRF-2 and GR genes were elevated in the treated mice. However, statistical analysis revealed that KRGE-loaded PLGA NPs were more effective than KRGE.Conclusion: Our results are consistent with the beneficial effect of KRGE on the treatment of PCOS. Nonetheless, KRGE coated with PLGA NPs were suggested more effective according to our findings.
