بررسی اثر درمانی ترکیب امواج فراصوت و داروی شیمی‌درمانی داکاربازین بر روی رده‌ی سلولی ملانوما

چکیده

مقدمه: استفاده از امواج فراصوت در ترکیب با داروهای شیمی‌درمانی، باعث بالا رفتن اثربخشی دارو در دوزهای کم می‌شود. این امر می‌تواند در بهبود سرطان‌های مقاوم به درمان مثل ملانوما نقش بسزایی ایفا کند. همچنین در این روش، پایین آمدن دوز دارو، سمیت ایجاد شده را بر سلول‌های سالم به حداقل می‌رساند. این پژوهش، با هدف بررسی تأثیر درمان ترکیبی امواج فراصوت و داروی شیمی‌درمانی داکاربازین بر میزان مرگ سلولی در ملانوما انجام شد.
روش‌ها: بعد از کشت سلول‌های ملانوما B16F10 به صورت آزمایشگاهی، غلظت بهینه‌ی داکاربازین به کمک تست MTT مشخص شد. همچنین تأثیر امواج فراصوت بر بقاء سلول‌ها به صورت مجزا بررسی گردید. در مرحله‌ی بعد، سلول‌ها به کمک غلظت بهینه‌ی داکاربازین و امواج فراصوت با فرکانس یک مگاهرتز در شدت‌های 5/0، 1، 5/1 و 2 وات بر سانتی‌متر مربع به مدت 3 دقیقه در مد پیوسته درمان شدند و بعد از گذشت 24 ساعت اکوباسیون، میزان بقاء به کمک آزمون MTT و میزان آپوپتوز به کمک فلوسایتومتری بررسی شد.
یافته‌ها: درصد بقاء سلول‌های ملانوما درمان شده به صورت ترکیبی با داروی داکاربازین و امواج فراصوت در شدت 2 وات بر سانتی‌متر مربع، بیشترین کاهش را نشان داد. همچنین سطح آپوپتوز ایجاد شده در شدت 5/1 وات بر سانتی‌متر مربع در ترکیب با داکاربازین، افزایش معنی‌داری داشت.
نتیجه‌گیری: استفاده از امواج فراصوت در ترکیب با دارو‌ی داکاربازین، می‌تواند در بهبود پاسخ ملانوما به درمان با این دارو، نقش مؤثری ایفا کند و همچنین باعث کاهش سمیت عمومی برای سلول‌های غیر سرطانی شود.

Background: The use of chemotherapy drugs in combination with ultrasound exposure increases their synergic effect at lower doses. It can play an important role in the treatment of drug-resistant cancers, such as melanoma. 
Also, in this method, reducing the dose of the drug minimizes side effects on healthy cells. This study aimed to evaluate the effect of ultrasound-mediated chemotherapy with dacarbazine on malignant cell death and apoptosis induction in melanoma. 
Methods: After culturing B16F10 melanoma cells in-vitro, the optimal concentration of dacarbazine was determined by MTT assay. The effect of ultrasound on cell survival was also investigated separately by the same method. In the next step, the cells were treated with the optimal concentration of dacarbazine at intensities of 0.5, 1, 1.5 and 2 w/cm2 and were observed for 3 minutes. After 24 hours MTT assay was used to measure cell 
viability. The level of apoptosis induction by treatments was also measured using flow cytometry.
Findings: The survival rate of melanoma cells treated in combination with dacarbazine and ultrasound at 2 w/cm2showed the greatest decline. Also, the rate of apoptosis at 1.5 w/cm2
in combination with dacarbazine increased significantly.
Conclusion: The use of ultrasound in combination with dacarbazine can be effective in improving the response of melanoma to treatment and reduce the overall toxicity to non-cancerous cells.
