بررسی بیان ژن‌های 1 HMGA- و 2 HMGA- در نمونه‌های تومور و طبیعی (حاشیه‌ی تومور) سرطان معده در خوزستان

مقدمه: سرطان معده، یکی از شایع‌ترین بدخیمی‌ها در سراسر جهان است که منجر به مرگ و میر بالایی می‏شود. خانواده HMGA (گروه با تحرک بالا) از پروتئین‌های تنظیم‌کننده‌ی ساختار کروماتین تشکیل شده است و نقش مهمی در تومورزایی دارد. این مطالعه با هدف بررسی تغییرات بیان این پروتئین‌ها در جمعیت بیمار خوزستان، ایران انجام شد.
روش‌ها: در این مطالعه، در مجموع 60 نمونه بافت شامل 30 نمونه‌ی تومور سرطان معده و 30 نمونه‌ی غیر توموری (حاشیه تومور) از بانک بافت تومور بیمارستان امام خمینی (ره) اهواز تهیه شد. پس از استخراج RNA و ارزیابی کمیت و کیفیت آن با استفاده از دستگاه نانودراپ و الکتروفورز ژل آگارز، ارزیابی بیان از بیان ژن‌های HMGA-1 و HMGA-2 با تکنیک Real-time PCR انجام شد. تجزیه و تحلیل آماری با استفاده از نرم افزار GraphPad Prism 9.2.0.332 به همراه روش‌های آماری T-test و ANOVA انجام شد.
یافته‌ها: نتایج نشان داد که بیان نسبی ژن HMGA-1 در نمونه‌های تومور نسبت به نمونه‌های غیر توموری به طور معنی‌داری 17 برابر افزایش یافت (0/0001 = P). به طور مشابه، بیان ژن HMGA-2 در نمونه‌های تومور به میزان قابل توجهی 15 برابر نسبت به نمونه‌های طبیعی افزایش یافت (0/0038 = P).
نتیجه‌گیری: این مطالعه به نقش بالقوه‌ی HMGA-1 و HMGA-2 در توسعه‌ی سرطان معده در نمونه‌های تومور بیماران در استان خوزستان، ایران اشاره دارد. این یافته با تحقیقات قبلی که نقش پروتئین‌های HMGA را در سرطان‌های مختلف برجسته می‌کرد، مطابقت داشت و نقش بالقوه آنها را در تومورزایی نشان داد.

Investigating the Expression of HMGA-1 and HMGA-2 Genes in Tumor and Normal Samples (Tumor Margin) of Gastric Cancer in Khuzestan

Background: Stomach cancer is one of the most common malignancies worldwide, which leads to high mortality. The HMGA (high mobility group) family consists of chromatin structures that regulate proteins and play an meaningful role in tumorigenesis. This study was conducted to investigate the expression changes of these proteins in the patient population of Khuzestan, Iran.
Methods: In this survey, 60 tissue samples, including 30 gastric cancer tumor samples and 30 non-tumor samples (tumor margins), were prepared from the tumor tissue bank of Imam Khomeini Hospital (RA) in Ahvaz. After extracting RNA and evaluating its quantity and quality using a nanodrop device and agarose gel electrophoresis, the expression of HMGA-1 and HMGA-2 genes was assessed with a Real-time PCR technique. Statistical analysis was performed using t-test and ANOVA statistical methods.
Findings: The results showed that the relative expression of the HMGA-1 gene in tumor samples compared to non-tumor samples increased significantly by 17 times (P = 0.0001). Similarly, HMGA-2 gene expression in tumor samples significantly increased 15 times compared to ordinary samples (P = 0.0038).
Conclusion: This study points to the potential role of HMGA-1 and HMGA-2 in the development of gastric cancer in medical tumor samples in Khuzestan province, Iran. These findings align with previous research highlighting the roles of HMGA in various cancers, consistent with their potential role in tumorigenesis.
