بررسی اثر تجویز منیزیم بر پاسخ احترازی غیرفعال و عملکرد حافظه در موش‌های صحرایی دیابتی

چکیده

مقدمه:
وجود منیزیم برای متابولیسم قند لازم است؛ این ماده ممکن است باعث آزاد سازی و فعال شدن انسولین شود. افزایش مقدار گلوگز خون در بیماران دیابتی باعث کاهش منیزیم خون و کمبود آن در ادرار می‌شود .

روش ها:
در این مطالعه 48 موش سفید صحرایی نژاد NMRI در چهار گروه دوازده تایی شامل سالم شاهد، دیابتی شاهد، سالم تحت تیمار منیزیم و دیابتی تحت تیمار منیزیم با چهار هفته تجویز سولفات منیزیم به صورت خوراکی با غلظت g/lit 10 به صورت محلول در آب مورد بررسی قرار گرفتند. القای دیابت با تزریق درون صفاقی استروپتوزتوسین با دوز 60 میلی‌گرم بر کیلوگرم، قند خون با استفاده از کیت بیوشیمیایی و به روش آنزیماتیک– کالریمتریک و سنجش حافظه با استفاده از آزمون یادگیری احترازی غیرفعال در شاتل باکس انجام گرفت. داده‌های مورد بررسی از گروه‌ها با سنجش غلظت گلوکز سرم و وزن حیوان جمع‌آوری و مقایسه شد.

یافته ها:
تفاوت بین گروه‌های سالم شاهد و دیابتی شاهد در افزایش غلظت گلوکز با 0001/0 > P معنی‌دار نشان داد. از طرفی غلظت گلوکز گروه دیابتی تحت تیمار منیزیم بعد از چهار هفته مصرف خوراکی منیزیم با گروه دیابتی شاهد پس از چهار هفته با 0001/0 > P کاهش معنی‌داری نشان داد، اما این تغییر در حیوانات سالم تحت تیمار منیزیم مشاهده نشد. در ارزیابی عملکرد حافظه، میانگین تأخیر زمانی ورود به محیط تاریک در دستگاه شاتل باکس به عنوان مبنای عملکرد حافظه در بازه‌های زمانی 1، 6، 24 و 48 ساعت و 1 و 2 هفته پس از سومین مرحله یعنی آموزش همراه با شوک، در کل 6 بازه‌ی زمانی در بین دو گروه سالم شاهد و گروه دیابتی شاهد با 001/0 > P معنی‌دار بود؛ تفاوت این مقادیر بین گروه دیابتی تحت تیمار منیزیم و سالم شاهد معنی‌دار نبود.

نتیجه گیری:
نتایج حاکی از هم‌خوانی مناسبی بین درمان دیابت با منیزیم و به خصوص کاهش گلوکز سرم حیوانات دیابتی به حد طبیعی و نیز اصلاح عملکرد حافظه و به خاطر آوری بود. مصرف خوراکی و درازمدت منیزیم موجب افزایش توانایی حیوان برای ذخیره نمودن اطلاعات در انبارهای حافظه و افزایش قدرت به یاد آوری اطلاعات انبار شده در حیوانات دیابتی می‌گردد.

چکیده [English]

Background:
Magnesium is an essential metal in carbohydrate metabolism that causes activation and release of insulin. In diabetic patients, increasing blood sugar results to decrease of serum Magnesium and its low concentration in urine.

Methods:
In this study, 48 NMRI rats (Razi co. Iran) categorized in 4 groups; normal control, diabetic control, normal with magnesium treatment, and diabetic with oral magnesium treatment of 10 g/lit (MgSo4) in water. Diabetes was induced with inter peritoneal prescription of 60 mg/kg streptozotocin (STZ). Glucose concentration was measured with enzymatic-calorimetric method. The learning and memory capacities (acquisition and processing of information, decision making and response initiating functions) were assessed by the passive avoidance in a shuttle box. The differences among sessions in the number of shuttle-avoidance responses were interpreted as learning and memory. The latency time (LT) for enter to dark compartment in 4 groups was measured in 1, 6, 24, 48 hours, one week and two weeks after initial shock through learning test.

Findings:
The statistical difference of glucose concentrations between normal and diabetic control groups was significant (P < 0.0001). But there was no significant difference among normal control and magnesium treatment diabetic groups. Memory capacities and function in 6 timescales showed statistical difference of mean latency time between normal and diabetic control groups (P < 0.001). There was not significant difference in latency time between magnesium treatment diabetic and normal control groups.

Conclusion:
The reduction of glucose concentration in diabetic rats after 4 weeks oral treatment with magnesium show a good correlation with increasing their avoidance and latency time of enter to dark compartment. Thus magnesium consuming of diabetic group resulted to modulation of memory through acquisition and processing of information, decision making and response initiating functions.
