چکیده

مقدمه: کاندیدا ارتوپسیلوزیس و کاندیدا متاپسیلوزیس گونه‌‌ها‌ی مخمری تازه کشف ‌شده‌ای هستند که از لحاظ فنوتیپی از گونه‌ی کاندیدا پاراپسیلوزیس غیرقابل تشخیص می‌‌باشند. در مطالعه‌ی حاضر، با استفاده از روش‌‌ها‌ی مولکولی فراوانی و انتشار کاندیدا ارتوپسیلوزیس و کاندیدا متاپسیلوزیس در بین جدایه‌ها‌ی کاندیدا پاراپسیلوزیس در نمونه‌‌ها‌ی مختلف بالینی در ‌ایران بررسی شده است.روش‌ها: DNA ژنومی ‌‌مخمری 935 جدایه‌ی مخمری با استفاده از فیلتر FTA از کشت تازه‌ استخراج شد. ابتدا ناحیه‌ی ITS مخمر‌ها‌ با پرایمر‌‌ها‌‌ی عمومی ‌‌ITS1 و ITS4 تقویت و پس از برش آنزیمی ‌‌باMspI  گروه کاندیدا پاراپسیلوزیس شناسایی شد. سپس ژن SADH تقویت و سه گونه‌ی کاندیدا پاراپسیلوزیس، متاپسیلوزیس و ارتوپسیلوزیس به کمک هضم آنزیمی ‌‌با NlaIII افتراق داده شد.یافته‌ها: در مجموع 111 جدایه به عنوان گروه پاراپسیلوزیس شناسایی شد که از بین آن‌ها‌ 18 جدایه (2/16 درصد) کاندیدا ارتوپسیلوزیس و 93 جدایه (7/83 درصد) کاندیدا پاراپسیلوزیس بودند. هیچ یک از جدایه‌‌ها‌‌ به عنوان کاندیدا متاپسیلوزیس شناسایی نشد. بیشترین جدایه‌‌ها‌‌ی کاندیدا ارتوپسیلوزیس مربوط به نمونه‌‌ها‌‌ی ناخن بود.نتیجه‌گیری: کاندیدا ارتوپسیلوزیس گونه‌ی کمیابی در‌ ایران نبوده و به نظر می‌رسد تعداد زیادی از استرین‌‌ها‌‌یی که ممکن است در ‌ایران به عنوان کاندیدا پاراپسیلوزیس شناسایی شوند، در واقع گونه‌ی ارتوپسیلوزیس باشند.

Background: Candida orthopsilosis and C. metapsilosis are recently introduced species that phenotipically indistinguishable from C. parapsilosis. In the present study, we evaluated the incidence and distribution of C. orthopsilosis and C. metapsilosis among 111 C. parapsilosis, isolated from different body locations by molecular methods.Methods: Yeast genomic DNAs were extracted from 935 isolates using Whatman FTA filter matrix technology from fresh cultures. First, ITS region of yeasts, were amplified by ITS1 and ITS4 primers, then Candida parapsilosis group were identified with digestion by MspI. SADH gene was amplified and three species of Candida including C. parapsilosis, C. orthopsilosis and C. metapsilosis were differentiated with digestion by NlaIII enzyme.Finding: Totally 111 isolates were identified as Candida parapsilosis among them, 18 isolates (16.2%) were identified as C. orthopsilosis and 93 isolates (83.7%) as C. parapsilosis. No isolates were identified as C. metapsilosis. The most isolates of C. orthopsilosis, were obtained from nail.Conclusion: C. ortopsilosis is not a rare species in Iran and it seems that a significant amounts of Candida strains which are considered as C. parapsilosis might be C. ortopsilosis.
