بررسی برون تنی عصاره‌ی هیدروالکلی سیر وحشی (Allium ampeloprasum) بر فعالیت حرکتی ایلئوم موش صحرایی

چکیده

مقدمه: در این تحقیق، اثر ضد انقباضی عصاره‌ی هیدروالکلی برگ گیاه تره‌ی کوهی بر انقباضات ایلئوم موش‌های صحرایی نژاد ویستار و مکانیسم محتمل بر آن بررسی شد.روش‌ها: در این تحقیق مداخله‌ای تعداد 48 سر موش صحرایی، از نژاد ویستار به طور تصادفی به شش گروه هشت‌تایی به شرح زیر تقسیم شدند: شاهد، دریافت کننده‌ی غلظت‌های تجمعی عصاره‌ی تره‌ی کوهی، دریافت کننده‌ی L-NAME (L-NG-nitroarginine methyl ester) و عصاره، نالوکسان و عصاره، پروپرانولول و عصاره و گروه دریافت کننده‌ی غلظت‌های تجمعی کلرور کلسیم. در روز آزمایش، ایلئوم موش‌ها جدا و انقباضات آن در حمام بافت تحت یک گرم کشش با اضافه کردن کلرور پتاسیم (mM 60) به روش ایزوتونیک ثبت شد. جهت بررسی مکانیسم اثر عصاره، بافت با پروپرانول، نالوکسان و یا L-NAME انکوبه شد و درصد تغییرات نیروی انقباضی بر روی کاغذ ثبت و محاسبه گردید. همچنین جهت نقش کانال‌های کلسیم در فعالیت حرکتی بافت، ایلئوم تحت تأثیر غلظت‌های تجمعی کلرور کلسیم قرار داده شد.یافته‌ها: عصاره‌ی تجمعی تره‌ی کوهی (100، 200 و 400 میلی‌گرم بر کیلوگرم) به صورت وابسته به غلظت انقباضات ایلئوم به وسیله‌ی کلرور پتاسیم را کاهش داد (0001/0 > P). پروپرانولول اثر مهاری عصاره بر انقباضات ناشی از کلرور پتاسیم را به صورت معنی‌داری کاهش داد (0010/0 > P). اما، L-NAME و نالوکسان سبب کاهش اثر مهاری عصاره بر ایلئوم نشدند. همچنین کلسیم سبب انقباض بافت دپولاریزه شده توسط کلرور پتاسیم شد و این اثر انقباضی توسط غلظت‌های تجمعی عصاره به طور معنی‌داری کاهش یافت (0010/0 > P).نتیجه‌گیری: عصاره‌ی هیدروالکلی برگ تره‌ی کوهی توانست با اثر بر گیرنده‌های بتاآدرنرژیک و کانال‌های کلسیم وابسته به ولتاژ، بر فعالیت حرکتی ایلئوم موش صحرایی اثر بگذارد و با توجه به نتیجه‌ی اثر، ممکن است بتوان از آن در درمان ناراحتی‌های گوارش استفاده کرد.

Background: Ampeloprasum is traditionally used for gastrointestinal disorders. In this experimental study, the effect of ethanolic extract of Allium ampeloprasum was investigated on the rat ileum contractions. The mechanisms were also evaluated.Methods: In this experimental study, 48 male Wistar rats were randomly designated into six equal groups, including control and cumulative concentrations of Allium ampeloprasum, propranolol, naloxone, L-name, and CaCl2. In the day of the experiment, the ileum was dissected and its contractions were recorded in Tyrode solution in an organ bath. To evaluate the mechanisms involved, propranolol, naloxone, or L-name was incubated and the percent of changes in contractions were recorded.Findings: The extract of Allium ampeloprasum dose-dependently reduced the contractions induced by KCl (60 mM) (P < 0.0001); propranolol reversed this effect (P < 0.0010), but L-name and naloxone were not effective. CaCl2 increased the contractions (P < 0.0010) and the extract reversed this effect.Conclusion: Allium ampeloprasum leaves can affect the ileum contractions through beta adrenoreceptors and voltage-dependent calcium channels and might be used in gastrointestinal disorders.
