سمیت عصاره‌ی آبی- الکلی کلپوره (Teucrium polium) بر کلیه‌ی موش صحرایی

چکیده

مقدمه: گیاه کلپوره که به صورت گسترده‌ای در بین مردم استفاده می‌‌شود، می‌تواند دارای عوارض جانبی بر روی اعضای مختلف به ویژه کلیه باشد. در پژوهش حاضر، سمیت عصاره‌ی آبی- الکلی گیاه کلپوره بر کلیه‌‌ی موش صحرایی بررسی شد.روش‌ها: 100 موش صحرایی به طور تصادفی به شرح زیر به 10 گروه مساوی تقسیم شدند: گروه شاهد اول 28 روز بعد از تزریق داخل صفاقی آب مقطر و گروه شاهد دوم 28 روز بعد از آن کشته و نمونه‌گیری شدند. گروه‌های 3 الی 6 غلظت‌های mg/kg 200، 150، 100 و 50 عصاره را دریافت و 28 روز بعد کشته شدند. گروه‌های 7 الی 10 همین دوزها را دریافت کردند و روز 58 کشته شدند. بر اساس درصد واکولیزاسیون سلول توبولی، دیلاتاسیون و دژنراسیون سلول توبولی و کشت هیالین و وجود دبریس در سلول توبول، لزیون‌ها به نمره‌های 4-1 دسته‌بندی و گروه‌ها با یکدیگر مقایسه شدند.یافته‌ها: کلپوره به صورت وابسته به غلظت بر روی سلول‌های توبول کلیوی اثرات توکسیک داشت (020/0 = P). به علاوه، پس از قطع آن به مدت 28 روز و آزاد نگه داشتن موش‌ها، توبولار توکسیسیتی که شامل فلاتنینگ، واکوئولیزاسیون و دژنرسانس توبولر سلولی بود نیز پا برجا ماند و یا به صورت تأخیری، تشدید شد (030/0 = P).نتیجه‌گیری: با توجه به اثرات توکسیک کلپوره بر کلیه توصیه می‌شود در مصرف گیاه کلپوره دقت زیادی به عمل آید؛ زیرا ممکن است در انسان نیز اثرات مشابهی داشته باشد.

Nephrotoxicity of Hydroalcoholic Extract of Teucrium Polium in Wistar Rats

Background: Teucrium polium L is widely used for different pathological conditions and may have side effects, especially on kidney. The aim of this study was to evaluate the nephrototoxicity of hydroalcoholic extract of Teucrium polium in male Wistar rats.Methods: In this experimental study, 100 rats were designated in 10 groups of 10. Five groups were injected intraperitoneally (IP) 50, 100, 150, or 200 mg /kg extracts or normal saline for 28 days and sacrificed to study the kidney degenerations. Five other groups were injected the same drug regimen, but they were sacrificed 28 days after cessation of drug injections to investigate the effect of possible complication or regeneration during recovery.Findings: The Teucrium polium consumption had dose-related toxic effects on kidney (P = 0.02). Following 28 days of Teucrium polium consumption (phase 1), kidney damages were not increased in comparison with control group (P > 0.05). However, following 28 days of drug cessation, kidney damages including degeneration, destruction and vacuolization were significantly appeared in comparison with control group and with increasing the doses of Teucrium polium (P = 0.03).Conclusion: Due to nephrotoxicity, Teucrium polium should not be used or should be consumed with great caution.
