چکیده
مقدمه: دیابت نوع 2 شایعترین بیماری متابولیکی مزمن است که نرخ شیوع آن به سرعت در حال افزایش است و تعداد افراد زیادی را مبتلا میکند. علت این بیماری، ناشناخته است و گفته میشود برهمکنش عوامل ژنتیک، اپیژنتیک و محیطی از جمله چاقی مستعد کنندهی ابتلا به بیماری دیابت نوع 2 هستند. miRNA به عنوان یکی از عوامل کلیدی در تنظیم بیان ژن با مهار ترجمه و یا تجزیهی mRNA عمل میکند و این احتمال وجود دارد که miRNAها در رخداد مقاومت انسولین و دیابت نوع 2 نقش داشته باشند. یافتههای جدید استفاده از سلولهای تک هستهای خون محیطی را به عنوان منعکس کنندهی وضعیت پاتولوژیک بافتها معرفی کردهاند که میتواند منبع مناسبی جهت مطالعات و تشخیصهای مولکولی در بیماریها همچون بیماری دیابت نوع 2 باشد. از جمله miRNAهای دخیل در مراحل مختلف دیابت نوع 2، به خصوص مقاومت انسولین به عنوان اولین و مهمترین عامل خطر دیابت نوع 2، 103-miR میباشد. هدف از این مطالعه، بررسی بیان نسبی 103-miR در سلولهای تک هستهای خون محیطی رتهای مبتلا به دیابت القا شده به واسطهی رژیم غذایی چرب/ تزریق Streptozotocin در دو مرحلهی پیش دیابت و دیابت و مقایسهی آن با رتهای سالم بود.روشها: مدل حیوانی دیابت نوع 2 با استفاده از ترکیب رژیم غذایی با چربی بالا و تزریق دوز پایین Streptozotocin (mg/kg 35) ایجاد شد. سلولهای تک هستهای خون محیطی رتهای مبتلا در دو مرحلهی پیش دیابت (قبل از تزریق Streptozotocin) و دیابت (پس از تزریق) و همچنین رتهای سالم جداسازی شد. RNA با استفاده از Trizol از سلولها استخراج و سپس به cDNA (Complementary DNA) تبدیل شد. سطح بیان 103-rno-miR به وسیلهی تکنیک qRT-PCR (Quantitative reverse transcriptase- polymerase chain reaction) و حضور معرف SYBR Green و با استفاده از پرایمرهای اختصاصی LNA TM 103- Primerset rno-miR سنجیده شد.یافتهها: ترکیب رژیم غذایی چرب همراه با تزریق دوز پایین STZ میتواند دیابت نوع 2 را به خوبی در رتها القا کند. میانگین بیان 103-rno-miR در رتها در مرحلهی دیابت و پیش دیابت نسبت به رتهای سالم تفاوت معنیداری (050/0 ≥ P) را نشان داد.نتیجهگیری: یافتههای پژوهش حاضر، اهمیت 103-miR را در پاتوژنز دیابت نوع 2 به خصوص در مراحل اولیه نشان میدهد و همچنین امکان مناسب بودن سلولهای تک هستهای خون محیطی را به عنوان یک منبع در دسترس و غیر تهاجمی در مطالعات و یا تشخیصهای مولکولی برای پیشبینی و پیگیری شرایط بیماری دیابت فراهم میکند.

چکیده English
Background: Type 2 diabetes (T2D) is the most prevalent chronic metabolic disease. MicroRNAs act as regulators of gene expression by inhibiting translation or promoting degradation of target mRNAs. Recent reports indicate that microRNAs can play important role in establishment of diabetes and has roles in type 2 diabetes different aspects including insulin insensitivity. To detect a noninvasive screening method for type 2 diabetes, we evaluated the expression level of miR-103 in peripheral blood mononuclear cells (PBMC) samples from pre-diabetes and type 2 diabetes rats in compare to normal group (n = 8 in each group) using real-time polymerase chain reaction (RT-PCR) method.Methods: Animal model type 2 diabetes was made with combination of high-fat diet (HFD) and low dose of stretozotocin (STZ) injection. Blood glucose level, serum insulin, and cholesterol, low density lipid and triglyceride levels were measured after 10-weeks diet interference. Peripheral blood mononuclear cells isolated from blood samples of three groups of animals, and then total RNA was obtained using Trizol reagent. All microRNAs converted to cDNA (complementary DNA). Expression level of rno-miR-103 was assessed via one-step SYBR GreenⅠ relative real-time polymerase chain reaction, using specific LNA TM qRT-PCR primer set for amplifying rno-miR-103. Statistical analysis was performed using SPSS20 software.Findings: Combination of high-fat diet and low dose of stretozotocin effectively induced type 2 diabetes in rats. rno-miR-103 expression level in peripheral blood mononuclear cells samples of pre-diabetic (high-fat diet) subjects and type 2 diabetes rats significantly elevated in compare to normal group.Conclusion: The high-fat diet/stretozotocin-induced type 2 diabetes animal model is the most suitable way for mimicking pre-diabetes and diabetes in men. The result of miRNA expression assay in peripheral blood mononuclear cells indicates the importance of rno-miR-103 in type 2 diabetes pathogenesis in early stages and has potential as a clinically useful noninvasive source to predict the disease status.
