بررسی اثر دود حاصل از عصاره‌ی مستقیم عنبرنسارا بر باکتری‌های گرم مثبت Staphylococcus aureus و Bacillus subtilis

چکیده

مقدمه: استفاده از دودهای طبی جایگزین در درمان بیماری‌ها و از جمله بیماری‌های میکروبی و عفونی در ایران از سال‌های دور رواج داشته است. این مطالعه، با هدف مقایسه‌ی خواص ضد میکروبی دود حاصل از عصاره‌ی مستقیم عنبرنسارا با آنتی‌بیوتیک‌های رایج بر روی Staphylococcus aureus و Bacillus subtilis بود.روش‌ها: در این مطالعه‌ی آزمایشگاهی، دود عنبرنسارا به عنوان مداخله‌ای بر سوش‌های مورد و گروه‌های آنتی‌بیوتیک بر سوش‌های مشابه به عنوان شاهد در نظر گرفته شدند. سوش‌های استاندارد Staphylococcus aureus و Bacillus subtilis در محیط کشت‌های مناسب کشت داده شدند. عصاره‌گیری عنبرنسارا بدون استفاده از حلال انجام شد تا احتمال تأثیر حلال بر میکروب‌ها را از بین ببرد. سری رقت و آزمایش دیسک دیفیوژن برای هر کدام از باکتری‌ها سه بار انجام شد.یافته‌ها: میانگین هاله‌ی عدم رشد در Staphylococcus aureus به طور معنی‌داری بیشتر از ونکومایسین بود؛ اما میانگین هاله‌ی عدم رشد در Bacillus subtilis با سیپروفلوکساسین تفاوت معنی‌دار نداشت. آزمون تعقیبی Least significant difference (LSD) نشان داد که قطر هاله‌ی عدم رشد در Staphylococcus aureus به طور معنی‌داری بیشتر از Bacillus subtilis بود. در حالی که، Minimum inhibitory concentration (MIC) برای Bacillus subtilis بیشتر از Staphylococcus aureus و Minimum bactericidal concentration (MBC) برای Bacillus subtilis بیشتر از Staphylococcus aureus بود.نتیجه‌گیری: دود حاصل از عصاره‌ی عنبرنسارا بر رشد Staphylococcus aureus تأثیر معنی‌داری داشت و می‌توان آن را به عنوان دارو پیشنهاد نمود.

چکیده [English]

Background: The use of medicinal smokes in treatment of many diseases, including infectious microbial diseases, has long been popular in Iran. This study aimed to compare the antimicrobial effects of smoke of Anbar Nasara (donkey dung) extract on Staphylococcus aureus and Bacillus subtilis with common antibiotics.Methods: In this experimental study, donkey dung smoke as medicine and antibiotics as a control group were considered. Standard strains of Staphylococcus aureus and Bacillus subtilis were cultured in suitable mediums. Anbar Nasara extract was prepared without use of solvent, in order to prevent the antibacterial effect of solvent on microbes. Dilution and disk diffusion test was performed 3 times for each of the bacteria.Findings: The mean of inhibition zone of Staphylococcus aureus by Anbar Nasara smoke was significantly greater than the inhibition zone induced by vancomycin; but there was no significant difference between the inhibition zone of Bacillus subtilis induced by Anbar Nasara smoke and ciprofloxacin. Least significant difference (LSD) post hoc test showed that the inhibition diameter of Staphylococcus aureus was significantly higher than the Bacillus subtilis. Bacillus subtilis induced higher minimum inhibitory concentration (MIC) values than Staphylococcus aureus; but, Bacillus subtilis induced higher minimum bactericidal concentration (MBC) values than Staphylococcus aureus.Conclusion: The smoke of Anbar Nasara extract has a significant inhibitory effect on Staphylococcus aureus growth; so it can be offered as a medicine.
