طراحی و کلون کردن microRNA-148b در شاتل وکتور لنتی ویروسی

چکیده

مقدمه: ریزRNAها (miRNAs)، RNAهای غیر کد کننده‌ی کوچکی هستند که بیان ژن را از طریق اتصال به mRNA هدف تنظیم می‌نمایند. از آن‌جایی که این مولکول‌ها در تکثیر و تمایز سلولی نقش مهمی دارند، می‌توان از آن‌ها در پزشکی ترمیمی استفاده کرد. از میان حاملین مختلفی که برای رها‌سازی این الیگونوکلئوتیدها وجود دارد، سیستم لنتی ویروسی حامل مناسبی به نظر می‌رسد. از طرف دیگر، مشخص شده است که miR-148b در استئوژنز نقش دارد. بدین منظور، در مطالعه حاضر طراحی و کلون کردن microRNA-148b در شاتل وکتور لنتی ویروسی مورد بررسی قرار گرفت.روش‌ها: ابتدا پرایمرهای رشته‌های p3 و p5 microRNA-148b طراحی گردید و پس از هیبریداسیون، داخل پلاسمید شاتل لنتی ویروسی کلون شد. سپس ورود صحیح قطعه‌ها با استفاده از هضم آنزیمی و توالی‌یابی مورد بررسی قرار گرفت. در ادامه، پس از تولید ذرات لنتی ویروسی مربوط، از آن‌ها برای ترانس‌داکشن سلول‌های بنیادی مزانشیمی استفاده گردید.یافته‌ها: miR-148b-3p/-5p طراحی شده با موفقیت داخل شاتل لنتی ویروسی کلون گردید. یافته‌های حاصل از هضم آنزیمی و توالی‌یابی، مؤید ورود موفقیت‌آمیز رشته‌های مورد نظر داخل پلاسمید لنتی ویروسی بود. بیان بالای Enhanced Green Fluorescent Protein (eGFP) نشان دهنده‌ی کارایی بالای ترانس‌فکشن و ترانس‌داکشن بود.نتیجه‌گیری: در مطالعه‌ی حاضر دو نوع ویروس حامل miR-148b-3p و miR-148b-5p تولید گردید که می‌توانند جهت بررسی فرایند استئوژنز مورد بررسی قرار گیرند.

چکیده [English]

Background: Micro (mi)RNAs are non-coding endogenous RNAs which regulate gene expression by hybridization to specific binding sites in target mRNA sequences. Since several miRNAs are involved in proliferation and differentiation, miRNA-based therapies could be promising approach in regenerative medicine. Among different vehicles, lentiviral vector system is suitable for miRNA delivery. Besides, it is shown that miRNA-148b is involved in osteogenic differentiation. In this study, designing and cloning of miR-148b to lentiviral vector were investigated.Methods: We introduced miRNA-148b-3p/-5p into lentiviral vector through cloning producers. The sequences of lentiviral vectors carrying miRNA-148b were checked via analytical digestion as well as Sanger DNA sequencing. In the following, produced lentiviral vectors were used for mesenchymal stem cells transduction.Findings: Designed miR-148b-3p/-5p successfully cloned to the shuttle. Correctness and absence of any unintended mutations of lentiviral shuttle carrying miRNA-148b3p/-5p were confirmed followed by lentiviral production. Expression of enhanced green fluorescent protein (eGFP) demonstrated high efficiency of transfection as well as transduction.Conclusion: Viral vectors constructed in this study could be used for investigation of osteogenesis.
