مقدمه: ملانوما، یکی از کشندهترین سرطانهای پوست میباشد که اثربخشی درمانهای رایج بر روی آن، چشمگیر نمیباشد. پلاسمای اتمسفری سرد، راهکاری جدید در درمان سرطان محسوب میشود. این مطالعه با هدف ارزیابی اثر فوتوداینامیک پلاسمای سرد در حضور حساسکنندهی نوری ایندوسیانین گرین بر ردهی سلولی ملانوما انجام شد.
روشها: در این مطالعه، از دستگاه پلاسما با گاز هلیوم در حضور ایندوسیانین گرین جهت تیمار ردهی سلولی ملانوما (DFW) استفاده شد. بدین منظور، ابتدا غلظت بهینهی ایندوسیانین گرین پس از 24 ساعت انکوباسیون با استفاده از تست MTT مشخص گردید. سپس سلولها با و بدون حضور ایندوسیانین گرین تحت تابش پلاسمای سرد در زمانهای مختلف 15، 30، 60 و 90 ثانیه قرار گرفتند. درصد بقای سلولها 48 ساعت بعد از درمان با پلاسمای سرد با استفاده از آزمون MTT ارزیابی شدند.
یافتهها: درمان پلاسمای سرد اثر سمیتی وابسته به زمان ایجاد میکند. درصد بقای سلولهای تیمار شده با پلاسمای سرد در حضور ایندوسیانین گرین به طور معنیداری نسبت به سلولهای تیمار نشده کاهش یافت. بیشترین کاهش درصد بقا در گروه درمانی، 90 ثانیه پلاسمای سرد و غلظت 20 میکرومولار (Indocyanine green) ICG مشاهده گردید.
نتیجهگیری: مطالعهی حاضر، خاصیت ضدسرطانی پلاسمای اتمسفری سرد را در درمان سلولهای ملانوما تأیید کرد. همچنین نشان داد که پلاسما میتواند جایگزین مناسبی در درمان فوتوداینامیک باشد و ایندوسیانین گرین حساسکنندهی نوری مناسبی در درمان با پلاسما خواهد بود.

Background: Melanoma is one of the deadliest types of skin cancers. The efficacy of current treatments is not significant. Cold atmospheric plasma (CAP) is a new modality for cancer treatment. This study aimed to evaluate the photodynamic effects of Indocyanine green in cold plasma on melanoma cell line.
Methods: In this study, helium based cold plasma and Indocyanine green were used to treat DFW cells. In this respect, at first the optimal concentration of Indocyanine green was determined by MTT test after an overnight incubation. Then cells were treated to cold plasma with and without Indocyanine green in different times such as 15, 30, 60 and 90 s. The cell survival rate was evaluated by MTT test 48 h after CAP treatment.
Findings: Cold plasma treatment produces cytotoxic cell with a time dependent decline. The survival rate was significantly reduced in treated cells to cold plasma and Indocyanine green compared to untreated cells. The viability decreased significantly in the cells treated to cold plasma. The most reduction in survival rate was observed in treated cells to cold plasma for 90 s and Indocyanine green with 20µM concentration.
Conclusion: This study approved anti-cancer effect of cold plasma on melanoma cells. In addition, it showed that CAP can be a good alternative to photodynamic therapy and Indocyanine green also a great photosensitizer at cold plasma treatment.
